Antidepressant-like activity of the neuropeptide Y Y5 receptor antagonist Lu AA33810: behavioral, molecular, and immunohistochemical evidence

Abstract: RationaleIt has recently been found that chronic treatment with the highly selective, brain-penetrating Y5 receptor antagonist, Lu AA33810 [N-[[trans-4-[(4,5-dihydro [1] benzothiepino[5,4-d] thiazol-2-yl) amino] cyclohexyl]methyl]-methanesulfonamide], produces antidepressant-like effects in the rat chronic mild stress model.ObjectiveIn the present study, we investigated the possible antidepressant-like activity of Lu AA33810 in rats subjected to glial ablation in the prefrontal cortex (PFC) by the gliotoxin L-… Show more

“…LP-NPY (Y1R and Y5R agonists) is as useful as NPY and presents therapeutic potential in preventing the development of depressive-like behavior ( 142 ). Nevertheless, Y5R antagonists also play an antidepressant role through the MAPK/ERK and PI3K signaling pathways ( 143 ). Besides, Y2R plays an antidepressant role by inhibiting the NLRP3 signaling pathway in LPS-induced depression model rats ( 87 ).…”

“…These results from this study show that TTK attenuated the depressive-like symptoms produced by the astroglial degeneration model of depression. After the glial ablation model of depression was developed [ 18 ], the antidepressant-like effect of mGluR5 antagonist MTEP [ 38 ], prefrontal cortex deep brain stimulation [ 39 ], Y5 receptor antagonist Lu AA33810 [ 40 ], Harmine [ 41 ], ZL006 [ 42 ], and ginsenoside Rf [ 43 ] were tested. In a manner similar to a previous study, infusion of L-AAA induced depressive-like behavior, and administration of imipramine [ 38 ] reversed the depressive behavior and led to a decrease in the number of GFAP-positive cells.…”

“…Among five NPY receptor subtypes, Y1R has been shown to mediate NPY-induced antidepressant-like activity in the olfactory bulbectomized rats and the PTSD model rats as well as in the forced swimming test mouse [4345]. Meanwhile, antagonist of Y5R has antidepressant-like effects in the CMS rat [46, 47]. In the present study, mRNA expression levels of Y1R, Y2R, and Y5R were measured in both the mPFC and the hippocampus of LPS rats; decreased Y2R expression was only found in the mPFC selectively.…”
