Antidiabetic and Antihyperlipidemic Effects of Clitocybe nuda on Glucose Transporter 4 and AMP‐Activated Protein Kinase Phosphorylation in High‐Fat‐Fed Mice

Abstract: The objective of this study was to evaluate the antihyperlipidemic and antihyperglycemic effects and mechanism of the extract of Clitocybe nuda (CNE), in high-fat- (HF-) fed mice. C57BL/6J was randomly divided into two groups: the control (CON) group was fed with a low-fat diet, whereas the experimental group was fed with a HF diet for 8 weeks. Then, the HF group was subdivided into five groups and was given orally CNE (including C1: 0.2, C2: 0.5, and C3: 1.0 g/kg/day extracts) or rosiglitazone (Rosi) or vehic… Show more

“…The experiment was determined using a procedure described elsewhere [ 20 , 21 ]. Total RNA was reverse transcribed to cDNA in a reaction mixture containing buffer, dNTP, the oligo (dT) primer, and Moloney murine leukemia virus reverse transcriptase and then heated at 90°C for 5 min to terminate the reaction.…”

“…In mice with Streptozotocin (STZ)-induced liver damage (Box 3), L. nuda aqueous extract, as an adjuvant to an 8-week HFD diet, induced a reduction in IHTG content that was similar to the positive control group, treated with rosiglitazone, a PPAR-agonist antidiabetic drug. This extract induced an upregulation of PPAR-α mRNA levels and an increase in p-AMPK, paralleled by a decrease in the hepatic expression of gluconeogenic genes and an increase in GLUT-4 protein levels in liver and muscle tissue respectively [209]. Ganoderma lucidum ethanol extract also increased p-AMPK and p-ACC (inactive form) protein content in both cellular models (HepG2 and 3T3-L1) and HFD-fed mice after 16 weeks of feeding [210].…”

“…AMPK regulates the phosphorylation of PFK2 [ 37 ]. AMPK also can inhibit the activation of FAS, ACC, etc., to inhibit gluconeogenesis and glycogen formation [ 38 , 39 ]. AMPK participates in the regulation of lipid metabolism, which is related to SREBP1C [ 40 ].…”
