The use of medicinal plants in traditional medicine is a common practice in developing countries. However, this unregulated or poorly rational use may present a dose-dependent risk of toxicity to humans. This study aimed to explore the phytochemical and toxicological characteristics of ten (10) plant species used in the traditional treatment of infectious diarrhea in Benin. The acute toxicity of aqueous and hydroethanolic extracts of Khaya senegalensis, Daniellia oliveri, Rauvolfia vomitoria, Vernonia amygdalina, Manihot esculenta, Ocimum gratissimum, Senna italica, Diospyros mespiliformis, Pterocarpus erinaceus, and Anacardium occidentale was evaluated following the OECD 423 protocol at a single dose of 2000 mg/kg. This safety test was complemented by a larval cytotoxicity test. Hematological and biochemical examinations, as well as a histological study of the liver and kidneys, were performed. Larval cytotoxicity was assessed by the sensitivity of Artemia salina larvae to different concentrations of the plant extracts studied. Testing for chemical compounds was performed on the basis of differential staining and precipitation reactions. The mean lethal concentration (LC50) was determined by the probit method. The qualitative phytochemical screening of the plants studied revealed the presence of catechic tannins, gallic tannins, flavonoids, anthocyanins and sterol-terpenes, alkaloids, saponosides, and reducing compounds. This composition varied according to the plants studied. Acute toxicity data indicated that there was no mortality and no structural and functional alterations of the liver and kidneys of treated animals. Larval cytotoxicity data suggest that the plants studied are not cytotoxic (LC50 ≥ 0.1 mg/mL). These observations reflect the safety of these plants and justify their use in traditional medicine in the treatment of many diseases including diarrheal diseases.

Background. People with diabetes, herbalists, and traditional medicine practitioners (TMPs) from Kinshasa use plants to treat diabetes, but no study has inventoried the plants used by these populations. The present study was conducted to identify the plants used in Kinshasa to treat diabetes mellitus. Methods. The survey conducted in the form of a semistructured interview between March 2005 and August 2006 made it possible to collect ethnobotanical information from people with diabetes (n = 126), herbalists (n = 80), and TMPs (n = 120). Results. The 326 subjects consulted (sex ratio M/F = 0.6, age 51 ± 7 years, and experience: 17 ± 5 years) provided information on 71 plants, most of which are trees (35%), belonging to 38 families dominated by Fabaceae (19.7%) and indicated in 51 other cases of consultation dominated by malaria (12%). From these 71 plants derived, 86 antidiabetic recipes were administered orally, where the leaf is the most used part (>50%) and the decoction (>46%) is the most common mode of preparation. This study reports for the first time the antidiabetic use of 11 species, among which Tephrosia vogeliiX (0.08), Chromolaena corymbosaX (0.06), and Baphia capparidifoliaX (0.06) present the highest consensus indexes (CI) and Marsdenia latifoliaW (UVp = 0.08) and Rauvolfia manniiX (UVp = 0.06) present the highest UVs. Conclusion. The results show that Kinshasa people treat diabetes using several plants. Some are specific to the ecological environment; others are used in other regions. Pharmacological studies are underway to assess the therapeutic efficacy of these plants.

In vivo antiplasmodial activity of the methanol leaf extract of Piliostigma reticulatum (Dc.) Hochst (Fabaceae)

Background Piliostigma reticulatum is a plant traditionally used to treat malaria, smallpox, neuralgia, dysentery, diarrhea, and rheumatism in Northern Nigeria. There is no scientific evidence to support the antimalarial activity of this plant. This work aims to investigate the in vivo antiplasmodial activity of the methanol leaf extract of Piliostigma reticulatum (MPR) in mice, infected with NK65 chloroquine-sensitive Plasmodium berghei. The oral lethal doses and preliminary phytochemical screening of the extract were performed. The therapeutic, suppressive, and prophylactic models were used for the antiplasmodial activity at the doses of 250, 500, and 1000 mg/kg of the MPR extract. Chloroquine and artesunate were used as the positive control drugs, while distilled water was used for the negative control group. The antiplasmodial activity was determined by comparing the mean parasite clearance in the treated groups, to the negative control group. Also the effect of the extract on the blood packed-cell volume of mice (PCV) was determined. Results The LD50 of MPR was found to be > 5000 mg/kg. Glycosides, saponins, tannins, flavonoids, triterpenes and alkaloids were the phytochemicals identified in the extract. The extract of MPR produced a significant reduction in the mean parasitemia level compared to the negative control in the curative test: MPR 250 (68.31%, P < 0.001), MPR 500 (76.53%, P < 0.001), and MPR1000 (83.65%, P < 0.001). The extract prolonged the survival of infected mice (18.8 days), compared to the negative control (5.2 days). The extract produced significant chemosuppression compared to the negative control; MPR 250 (73.79%, P < 0.001), MPR 500 (81.33%, P < 0.001), and MPR 1000 (78.37%, P < 0.001). The extract produced significant chemoprophylaxis compared to the negative control; MPR 250 (68.5%, P < 0.001), MPR 500 (58.7%, P < 0.001), and MPR 1000 (84.77%, P < 0.001). The extract was found to have no significant effect on the blood PCV of the treated groups compared to the negative control. Conclusions The study showed that the MPR extract has significant antiplasmodial activity in mice at the doses tested, and could justify the traditional use of the plant in the treatment of malaria in Northern Nigeria.
