Antifibrotic and Anti-Inflammatory Actions of α-Melanocytic Hormone: New Roles for an Old Player

Abstract: The melanocortin system encompasses melanocortin peptides, five receptors, and two endogenous antagonists. Besides pigmentary effects generated by α-Melanocytic Hormone (α-MSH), new physiologic roles in sexual activity, exocrine secretion, energy homeostasis, as well as immunomodulatory actions, exerted by melanocortins, have been described recently. Among the most common and burdensome consequences of chronic inflammation is the development of fibrosis. Depending on the regenerative capacity of the affected t… Show more

“…The α-MSH, an endogenous neuropeptide derived from POMC, is widely expressed in various tissues and organs and plays an important role in a variety of biological processes, such as energy metabolism, body weight regulation, sexual activity, and exocrine secretion ( 39 ). This hormone also has broad anti-inflammatory effects ( 40 ).…”

“…In dermal fibroblasts, α-MSH modulates collagen metabolism by improving the orientation of the collagen fibers [ 158 , 159 , 160 ]. α-MSH may drive the healing into a more regenerative/less scarring pathway by counteraction the pro-fibrotic action of TGFβ [ 160 , 161 ]. Recently, a higher prevalence of coarse collagen, pixel, and vessel density in photo-exposed areas has been associated with MC1R polymorphic variants [ 162 ].…”

“…α-MSH is also thought to inhibit NF-κB in T lymphocytes, leading to a decrease in IFN-γ production, whilst triggering upregulation of the anti-inflammatory cytokine IL-10 and Transforming growth factor (TGF)-β, possibly through cAMP responsive element binding protein (CREB). Thus, modulation of cAMP signaling by α-MSH can convert effector T lymphocytes into regulatory T lymphocytes ( 212 , 217 , 218 ). Furthermore, α-MSH was shown to also upregulate the production of IL-10 in macrophages and monocytes, which might assist the expansion of CM-initiating cells ( 216 , 219 ).…”
