Antifibrotic cardioprotection of berberine via downregulating myocardial IGF-1 receptor-regulated MMP-2/MMP-9 expression in diabetic rats

Abstract: Diabetic cardiac fibrosis increases ventricular stiffness and facilitates the occurrence of diastolic dysfunction. Our previous studies have shown that berberine, a natural alkaloid, attenuates cardiac ischemia-reperfusion injury in diabetic rats. The aim of present study was to investigate the effects of long-term berberine treatment on cardiac remodeling in diabetic rats and the underlying mechanisms. Diabetic rats induced by low-dose streptozotocin injection combined with 8 wk of high-fat diet displayed sig… Show more

“…In STZ-induced diabetic models (Table 4), berberine reduced total cholesterol, triglyceride and LDL-C, while increasing high-density lipoprotein cholesterol (HDL-C) [58,59,62,75,105,108,120,129,141]. In ApoE -/mice fed with high-fat diet (HFD), a model for Western diet-induced atherosclerosis, berberine significantly attenuated the development of severe cardiovascular symptoms [130].…”

“…BBR reduced the incidence and duration of arrhythmias induced by myocardial infarction (Wang et al, 2011). In addition, Li et al reported that BBR alleviates cardiac ischemic and reperfusion injury and cardiac remodeling in diabetic rats (Li et al, 2018). Dong et al (2018) found that berberine attenuates cardiac dysfunction by decreasing myocardial lipidemic profiles in diabetic cardiomyopathy.…”

“…Numerous studies have suggested a potential effect of berberine on lipid and glucose homeostasis, insulin sensitivity and insulin secretion, as well as antioxidant and anti-inflammatory effects in the treatment of diabetes mellitus (Cheng et al, 2006;Chueh and Lin, 2012;Yang et al, 2012;Li et al, 2014). In addition, Li et al reported that BBR alleviates cardiac ischemic and reperfusion injury and cardiac remodeling in diabetic rats (Li et al, 2018). “…Similar to previous studies, we found that CV treatment ameliorated cardiac dysfunction in DM rats, as evidenced by the increased EF%, FS%, and structural abnormalities. The MMPs, especially MMP‐2 and MMP‐9, were induced by HG treatment in CFs and contribute to the progression of DCM by function in excessive collagen deposition (Li et al, ; Meng, Uzui, Guo, & Tada, ; Shen et al, ). In our present, the results also showed that DM/HG treatment could induce the levels of CTGF, FN, MMP‐2, MMP‐9, Col‐I, and Col‐III.…”
