Antifungal effect of 5‐aminolevulinic acid PDT in Trichophyton rubrum

Abstract: In the present investigation, we have shown for the first time that the onychomycosis-inducing dermatophyte Trichophyton rubrum was able to metabolize 5-aminolevulinic acid (ALA) to protoporphyrin IX (PpIX) in liquid culture medium. We have established and optimized the culture conditions and could show the typical PpIX-induced red fluorescence which was evaluated qualitatively by Wood's light examination and fluorescent microscopic analysis. The optimum concentration of ALA was in the range of 1-10 mmol l(-1)… Show more

“…Recently, the antimicrobial efficacy of PDT with endogenous porphyrins was highlighted in reports that found it to be effective against bacteria, virus, fungi, and protozoa [5,[12][13][14][15][16][17]. Thus, in this study, we attempted to treat recalcitrant Malassezia folliculitis with MAL-PDT.…”

“…Although the currently FDA-approved PDT agent ALA has demonstrated an antifungal effect in T. rubrum in vitro, its limitation is the need for bioconversion to PpIX (38) prior to irradiation. Unlike ALA, Pc 4 does not require the bioconversion step.…”

“…Lately, several reports show the positive effect of PDT against bacteria, viruses, fungi and protozoa (3)(4)(5)(6). The major drawbacks of topical PDT are shallow penetration depth (<2 mm) of ALA and MAL and pain experienced during light exposure (2,(7)(8)(9).…”
