Antigenic heterogeneity of the reticular meshwork in the white pulp of mouse spleen

Abstract: Monoclonal antibodies against cellular components of reticular meshworks were produced by immunizing rats with heterogeneous stromal-cell population of mouse spleen. Immunohistochemical screening selected two antibodies, WP-1 and RPSC-2. WP-1 proved to immunostain the meshwork of the B area densely, leaving the marginal zone unstained; it also reacted sparsely with the meshwork of the T-cell region. In contrast, RPSC-2 selectively immunostained the meshwork of the T region. Immuno-electron microscopy clearly v… Show more

“…B lymphocytes were present at the periphery of the T lymphocyte accumulations. Bar 100 μm lymphocyte areas of the WP (Chilosi et al 1993;Satoh et al 1997;Van den Berg et al 1989;Yoshida et al 1991). The antigenic heterogeneity of the framework may induce the segregation of the lymphoid subclass (Satoh et al 1997;Yoshida et al 1991).…”

“…A heterogeneous population of reticulum cells has been observed immunohistochemically in the WP of rat and mouse spleen (Van den Berg et al 1989;Yoshida et al 1991). The distribution of extracellular matrix protein in the reticular framework is also heterogeneous in the T-and Blymphocyte areas of the WP (Chilosi et al 1993).…”

“…As naïve T cells migrate along the stromal cell-lined corridors, they are likely to have to migrate over the associated DCs, ensuring extensive DC-T cell contact. It is interesting to note that lymphoid follicles contain stromal cells that are ultrastructurally similar to the cells in T zones (27,28). Recent studies have provided evidence that many of the follicular stromal cells produce the B cell attractant, BLC (29,30).…”

“…In contrast, reticular cells of the spleen display an enzyme and an antigenic heterogeneity demonstrated by the use of different antibodies identifying subsets of fibroblastic reticular cells in mouse and rat (Müller-Hermelink et al 1974;Van den Berg et al 1989;Yoshida et al 1991; see Table 3 in Balogh et al 2004 for overview). Several antibodies distinguish between reticular cells of T-and B-cell areas, suggesting that the antigenic diversity may depend upon the contact with lymphocytes.…”
