RESUMO: "Potencial anti-inflamatório e analgésico do extrato hidrolisado deUnitermos: Agave sisalana, Asparagaceae, Agavoideae, hemolítica anti-inflamatória, analgesia. ABSTRACT:The hemolytic, anti-inflammatory and antinociceptive properties from hydrolyzed extract Agave sisalana Perrine ex Engelm., Asparagaceae (HEAS) was evaluated on classic inflammation models. Male Swiss mice and male Wistars rats received HEAS (500 mg/kg) in two administration p.o. and i.p. in saline solution 0.9%. The acid hydrolysis inhibited the hemolytic action of saponins due to the retreat of side chain sugar. The treatment of the ear induced oedema by xylene with HEAS significantly reduced in two routes 13±1.5 and 10±0.63 mg, respectively, p.o. and i.p., in comparison with controls 27±1.5 saline and 13.5±1.2 AAS. The HEAS also diminished edema induced by carrageenin 43±1.58 mg (p.o.) and 17±1.26 mg (i.p.), when compared with control groups 52±1.58 mg (saline) and 10.05±1.58 (indomethacin). HEAS showed analgesic effects in abdominal constrictions 30.7% (p.o.), 88.7% (i.p.) comparable to that produced by (AAS) 70.6%. However in granuloma cotton pellet a chronic model of inflammation just the i.p. pathway decreased granulomatous tissue (20.4±1.32 mg) compared with controls 30.5±2.53 mg (saline) and 20.2±2.18 mg (dexamethasone). These data suggest that HEAS has anti-inflammatory and analgesic activity on acute and chronic processes.

“…Destaca-se que na inflamação aguda duas fases são identificadas: a primeira, detectada em torno de uma hora, chamada de fase rápida, marcada pela liberação de histamina, serotonina e bradicinina; e a segunda, chamada de tardia (1-6 h), em que mediadores como as cininas e prostaglandinas são liberados após duas e três horas, respectivamente, as últimas resultando da ativação da ciclo-oxigenase 2 (COX-2) (Bispo et al, 2000;Silva et al,2005). Considerando que qualquer substância que cause inibição ou redução da ação da carragenina é considerada como tendo ação antiinflamatória (Kummer & Coelho, 2002), os resultados aqui encontrados demonstrando que o extrato aquoso da Amburana cearensis produziu redução do edema induzido pelo flogógeno a partir da segunda hora de administração, evidenciam que os constituintes ativos presentes nas sementes dessa espécie apresentam atividade antiinflamatória e que atuam na fase tardia da inflamação. Como visto (Tabela 2), o aumento da liberação da medula óssea de neutrófilos imaturos (bastões) para o sangue ocorre quando aumenta a demanda funcional de neutrófilos para os tecidos ou em casos de leucemias mielógenas ou mielomonocíticas agudas ou crônicas.…”

RESUMO:Foram avaliados os efeitos antiinflamatório, antibacteriano e mutagênico do extrato aquoso das sementes de Amburana cearensis. A atividade antiinflamatória foi avaliada em modelo de edema de pata induzido por carragenina, utilizando o extrato em concentrações de 10 % e 20 % nos grupos experimentais; AAS 10 mg/kg (v.o) no grupo padrão e água destilada no grupo controle. A atividade antimicrobiana foi determinada através do método de diluição em Agar, utilizando concentrações de extrato de 10 %, 7,5 %, 5 %, 2,5 % e 1 % em cepas de Sthaphylococcus aureus ATCC 27853, Escherichia coli ATCC 25922 e Pseudomonas aeruginosas ATCC 25923) e a atividade mutagênica foi determinada pelo teste de Allium cepa, utilizando extrato em concentrações de 0,02 mg/mL, 0,1 mg/mL e 0,5 mg/mL. O extrato aquoso das sementes de Amburana cearensis nas concentrações de 10 % e 20 % apresentou efeito antiedematogênico, estatisticamente significativo a partir de duas horas após administração do flogógeno, e tal efeito persistiu até 24 horas após a indução da resposta inflamatória. Quanto à atividade antibacteriana, o extrato não apresentou ação contra as cepas bacterianas de Sthaphylococcus aureus, Escherichia coli e Pseudomonas aeruginosas nas concentrações testadas. A análise dos resultados do teste de Allium cepa evidenciou ação tóxica (em concentração de 0,5 mg/mL) e mutagênica (micronúcleo 0,1 mg/mL e aberrações cromossômicas 0,1 mg/mL e 0,5 mg/mL) do extrato de Amburana cearensis em células meristemáticas de Allium cepa. Tais resultados sugerem potencial aplicação terapêutica no tratamento da inflamação. Contudo, também demonstram a necessidade de estudar para comprovar a segurança na utilização dessa espécie.Palavras chaves: Amburana cearensis. Planta medicinal. Edema. Antimicrobiano. The anti-inflammatory, antibacterial and mutagenic effects of the aqueous extract of Amburana cearensis seeds were evaluated. “…In a different study with health professionals, it was observed that 50% of respondents stated giving priority to hypertension during medical history; however, these same professionals have reported not knowing pharmacological changes between NSAIDs and antihypertensive drugs 14 . This is of paramount importance since the literature has already described the interactions between NSAIDs and selective COX-2 inhibitors, beta-blockers, diuretics and other anti-hypertensive drugs 1,2,19 . MÉTODO:Utilizou-se um questionário com 18 perguntas estruturadas, englobando aspectos demográficos, conhecimento atual, prática atual e a posição do profissional quanto ao uso das AINES. RESULTADOS: Foram avaliados 119 profissionais, destes, 84,9% cursaram a disciplina de Farmacologia. Os resultados indicam a necessidade de maior educação em relação à legislação corrente sobre a prescrição de fármacos, já que 47,8% afirmam aconselhar, 3,3% prescrever e 21% recomendar o uso de AINES, atos não permitidos para esses profissionais. CONCLUSÃO: Identificou-se fragilidade no conhecimento do profissional fisioterapeuta acerca da utilização do uso dos AINES, ressaltando-se a não identificação de reações adversas, interações farmacológicas e o conhecimento sobre a legislação. INTRODUCTIONThe search for knowledge update in different professional areas is mandatory today, especially in public health areas. Drugs have been object of concerns and several studies carried out worldwide in the medical area, emphasizing issues regarding their potential adverse reactions, prescription standards and the influence of advertising on their choice 1,2 . According to the Code of Professional Ethics in Physical Therapy and Occupational Therapy, approved by the Resolution Coffito-10, from July 03, 1978, physical therapists are forbidden to prescribe drugs. However, they should know how physical therapy may affect drug therapy and vice-versa 3 . Aiming at providing drug knowledge to these professionals, the pharmacology discipline is mandatory for Physical Therapy students in most Brazilian courses, being classically ministered for each class of drugs: pharmacodynamics, pharmacokinetics, major drugs, clinical use and side effects 4 . However, an Australian study has pointed to the need for better education of physical therapists about drugs, especially anti-inflammatory uses, indications, contraindications, pharmacological interactions and side effects 5 . Among drugs with excessive number of prescriptions there are non-steroid anti-inflammatory drugs (NSAIDs), with emphasis to the so-called selective cyclo-oxygenases-2 (COX-2) inhibitors 6 . Many NSAIDs have gone from the status of prescription drugs to METHOD:The study consisted of a questionnaire with 18 structured questions, encompassing demographic aspects, current understanding, current practice and professionals' position about the use of NSAIDs. RESULTS: We have evaluated 119 professionals, of whom 84.9% attended the discipline of Pharmacology. Results indicate the need for further education with regard to current legislation about drug prescription, since 47.8% stated advising, 3.3% prescribing and 21% recommending the use of NSAIDs, which is not allowed for these professionals. CONCLUSION: There is poor understanding of physical therapists with regard to the use of NSAIDs, especially the non identification of adverse reactions, drug interactions and knowledge of the legislation.
