Antineoplastic Agents, 99. Amaryllis belladonna

Abstract: Amaryllis belladonna bulbs were examined for constituents inhibitory against the murine P-388 lymphocytic leukemia (PS system). Two in vitro active alkaloids, acetylcaranine (2; 9PS ED50 0.23 microgram/ml) and ambelline (3; 9PS ED50 1.6 micrograms/ml), were isolated accompanied by undulatine. However, the non-chiral anhydrolycorinium chloride (5) was found to be the principal antineoplastic (3 PS, 64-69% life extension at dose levels 10 to 20 mg/kg in vivo, ED50 1.4 micrograms/ml in vitro) component. “…Anticancer, antimicrobial and anticholinesterase activities represent some of these biological properties [1]. The genus Amaryllis L. comprises two species, A. belladonna and A. acuminata , both native to southern Africa [2]. Also referred to by the folk names “belladonna-lily” and “naked-lady”, A. belladonna has been used for several centuries in the medicinal traditions of the Sotho, Xhosa and Zulu peoples of South Africa, and in Java for the treatment of “swelling” (a presumed synonym for cancer) [2,3].…”

“…1) As regards phytochemical studies on this plant, the isolation of phenanthridine alkaloids, fatty acids, sterols and triterpene alcohols have been previously reported. [2][3][4] Alkaloids isolated from the bulbs of the tribe Amaryllidaceae showed various pharmacological and microbiological effects, such as antiviral, 5) antimalarial, 6) cytotoxic [6][7][8][9] and antineoplastic activities, [10][11][12] as well as effects on diseases of the nervous system. 13) An EtOAc-soluble fraction of C. asiaticum var.…”
