Antinociceptive Effects of 14-Membered Cyclopeptide Alkaloids

Abstract: The analgesic potential of six 14-membered-ring cyclopeptide alkaloids, namely, franganine (1), discarine B (2), scutianines B (3), C (4), and D (5), and adouetine X (6), have been investigated. Among the compounds tested, only franganine (1) and adouetine X (6) produced antinociceptive effects in a mouse model of acute pain, without inducing undesirable side effects. Furthermore, compound 6 also exhibited a pronounced analgesic effect in a chronic neuropathic pain model in mice. It has been found that adoueti… Show more

“…The identification of the molecules was performed by high resolution mass spectrometry, 1D and 2D NMR experiments and comparison with spectroscopic data reported in the literature [15][16][17][18]. An additional purification by semi-preparative HPLC of an active fraction towards T. b. brucei afforded two new oleanolic acid ester derivatives: 3β-acetoxy-27-trans-caffeoyloxyolean-12-en-28-oic acid methyl ester (5) and 3β-acetoxy-27-cis-caffeoyloxyolean-12-en-28-oic acid methyl ester (6).…”

“…Generally, these peptides are the second metabolites of plant and about 455 cyclopeptides have been discovered in higher plants belonging to 26 families, 65 genera, and 120 species (Tan and Zhou 2006;Trevisan et al 2009). In particular, plants of Caryophyllaceae and Rhamnaceae families commonly contain cyclopeptides.…”

“…In Rhamnaceae probably Zizyphus genus (endemic to Indo-Pakistan region) is one of more worldwide studied species (Mahajan and Chopda 2009). Zizyphus nummularia is very important in ethnomedicinal system of Indo-Pakistan subcontinent and its bark is known to have antibacterial and analgesic activities (Shah et al 1990, Trevisan et al 2009). Discaria genus is a small shrub found in Southern lands of Argentina and Chile.…”
