Antioxidant Activity and Oxidative Stress-Oriented Apoptosis Pathway in Saccharides Supplemented Cryopreserved Sperm of Pacific Abalone, Haliotis discus hannai

Abstract: The Pacific abalone Haliotis discus hannai is a highly commercialized seafood in Southeast Asia. The aim of the present study was to determine the antioxidant activity and oxidative stress-oriented apoptosis pathway in saccharides supplemented cryopreserved sperm of Pacific abalone. Cryopreserved sperm showed impaired antioxidant defenses due to the reduced mRNA abundance of antioxidant genes (CAT, Cu/Zn-SOD, Mn-SOD, GPx, GR, and BCL-2), apoptosis inhibitor (HSP70, and HSP90) gene, and enzymatic antioxidant ac… Show more

“…This may have been influenced by an increase in LPO, thereby decreasing the sperm quality [ 33 ]. Moreover, decreased mRNA expression of antioxidant genes such as GPx and SOD might result in apoptosis [ 34 ], as shown by the higher number of sperm with membrane damage, acrosome loss, and broken DNA ( Table-2 ).…”

“…Then, the suspension was mixed with 2.5 mL thiobarbituric acid and heated at 100 °C for 15 min, followed by quickly cooling in an ice bath. After a 10 min centrifugation (5000 r/min), the absorbance of the supernatant was measured at 450, 532, and 600 nm, respectively [ 35 ].…”

“…Representative antioxidant enzymes operating in this context include superoxide dismutase (SOD) and catalase (CAT). SOD converts O 2 − to H 2 O 2 , which is further converted into nontoxic H 2 O and O 2 molecules by CAT (Hossen et al, 2022).…”
