The importance of medicinal plants for the treatment of different diseases is high from the aspects of the pharmaceutical industry and traditional healers. The present study involves nine different medicinal plants, namely, Neolamarckia cadamba, Nyctanthes arbor-tristis, Pogostemon benghalensis, Equisetum debile, Litsea monopetala, Spilanthes uliginosa, Desmostachya bipinnata, Mallotus philippensis, and Phoenix humilis, collected from Chitwan district of Nepal for biochemical analysis followed by the isolation of active plant fractions from the bioactive plant extract. The methanolic extracts of roots, barks, seeds, seed cover, and the other aerial parts of plants were used for the phytochemical analysis and biological activities. The DPPH (2,2-diphenyl-1-picrylhydrazyl) free radical scavenging assay was adopted to evaluate the antioxidant activity. Antibacterial activity was evaluated using the agar well diffusion method. The antidiabetic activity was studied by the α-amylase enzyme inhibition assay. The highest antioxidant activity was observed in extracts of Nyctanthes arbor-tristis followed by Mallotus philippensis (seed cover), Pogostemon benghalensis, Litsea monopetala, Phoenix humilis, and Neolamarckia cadamba with IC50 values of 27.38 ± 1.35, 32.08 ± 2.81, 32.75 ± 2.13, 33.82 ± 1.07, 40.14 ± 0.93, and 50.44 ± 3.75 µg/mL, respectively. The highest antidiabetic activity was observed in extracts of Phoenix humilis followed by Desmostachya bipinnata and Pogostemon benghalensis with IC50 values of 95.69 ± 6.97, 99.24 ± 12.6, and 106.3 ± 12.89 µg/mL, respectively. The mild α-amylase enzyme inhibition was found in extracts of Nyctanthes arbor-tristis, Spilanthes uliginosa Swartz, Litsea monopetala, and Equisetum debile showing IC50 values of 110.4 ± 7.78, 115.98 ± 10.24, 149.83 ± 8.3, and 196.45 ± 6.04 µg/mL, whereas Mallotus Philippensis (seed cover), Mallotus philippensis (seed), and Desmostachya bipinnata showed weak α-amylase inhibition with IC50 values of 208.87 ± 1.76, 215.41 ± 2.09, and 238.89 ± 9.27 µg/mL, respectively. The extract of Nyctanthes arbor-tristis showed high zones of inhibition against S. aureus (ATCC 25923) and E. coli (ATCC 25922) of ZOI 26 and 22 mm, respectively. The chemical constituents isolated from the active plant Nyctanthes arbor-tristis were subjected to GCMS analysis where the major chemical compounds were 11,14,17-eicosatrienoic acid and methyl ester. These results support the partial scientific validation for the traditional uses of these medicinal plants in the treatment of diabetes and infectious diseases by the people living in different communities of Chitwan, Nepal.

Effects of Nepal origin plant species of Vitex negundo, Catharanthus roseu, Aegle marmelos and Elaeocarpus ganitrus extracts on mild steel corrosion were explored in bioethanol (E100) and its blend (E15) in airtight condition at 25±2 °C using static immersion, inhibition efficiency and mechanism tests which were complemented with adsorption isotherms and potentiodynamic polarization studies. Corrosion resistance of the mild steel was increased with increasing 500-2000 ppm concentrations of each plant extract in E100 and E15 biofuels. Additions of V. negundo and C. roseus extract separately in both the biofuels seems to be more effective inhibition actions to prevent the mild steel corrosion than A. marmelos or E. ganitrus addition so as the corrosion rates of the mild steel in E100 and E15 are successfully lowered even than in commercial gasoline (E0). The results obtained from the corrosion rate revealed the order of the corrosion inhibition efficiency (IE) as V. negundo > C. roseus > A. marmelos > E. ganitrus. The maximum IE (IEmax) in V. negundo and C. roseus leaves was showed about 89-86% and 71-75%, respectively, at 2000 ppm concentration, in spite of the other two more plants leaf extract also used as the corrosion inhibitors for the mild steel in both E100 and E15 biofuels. The IE increased on increasing inhibitor concentration following the Langmuir and Temkin adsorption isotherms but decreased with immersion time which suggested that the corrosion inhibition mechanism is of physical type of adsorption of the leaves constituents on the mild steel surface. This study focused on the determination of secondary metabolites to an evaluation of the biological activity of Elaeocarpus variabilis Zmarzty fruit extract, an endemic medicinal plant. Their total Phenolic, tannin, flavonoids, alkaloids and saponins contents were quantified and their antioxidant activity, determined by DPPH, total antioxidant, ABTS, FRAP and Fe2+ chelating activity. The ethyl acetate extract exhibited the highest radical scavenging activity. The most relevant contents in total phenolics (Gallic Acid Equivalent mg/g), tannin (Gallic Acid Equivalent mg/g), total flavonoids (Rutin equivalent mg/g), total alkaloids (Quinine equivalent mg/g), and total saponin (Diosgenin equivalent mg/g) were observed. Significant correlations were established between antioxidant activity and secondary metabolites. Though the in vitro anti-diabetic activity was carried out using -amylase and -glucosidase inhibition, this indicates that the absorption and digestion of carbohydrates would probably reduce the blood glucose levels and hence help the management of type-2 diabetes. The ethyl acetate extract exhibited the potential of anticancer activity on human HT-29 colon cancer cell line IC50 values (30.0 ± 1.0) the valuable activity that compared to doxorubicin (17.0 ± 0.5). While this is the first study concerning extract from E. variabilis, our findings highlighted the richness’ in flavonoids of those extracts and their amazing potential as a source of antioxidant, anti-inflammatory, anti-diabetic, and anticancer agents.
