Hypericum scabrum (HS) widely used in traditional medicine due to its bioactive compounds was extracted by using ethanol-water (3:7, v/v). The extract was encapsulated with maltodextrin and gum arabic in a spray dryer in order to protect the phenolic compounds in its structure. Different amounts of microcapsules were added to our traditional drink, i.e. ayran (drinking yoghurt). The total phenolic content (TPC) and DPPH radical scavenging activity of the microcapsules in extract of HS and ayran samples were determined. The amount of total phenolic compounds in the microcapsule provided a superior effect than the extract. The ayran samples were supplemented with 2%, 3%, 4%, 5% and 6% of Hypericum scabrum microcapsules and it is observed that total phenolic content and DPPH radical scavenging activity indicated an increase with concentration. TPC and DPPH activity were determined as 268.86 mg GAE100 mL -1 and 78.05% for 6% supplemented samples. As a result of the sensory analysis, ayran samples supplemented with 4% of HS microcapsule gained the highest scores by the panelists and received more appreciation than the control group. It is concluded that HS4 (ayran produced by 4% HS supplemented microcapsule) sample was determined as the best sample according to the sensory analyses while the HS6 (ayran produced by 6% HS supplemented microcapsule) sample had the highest value in terms of DPPH scavenging activity and TPC results. The overall results of the present study revealed that 4% HS supplemented ayran can be produced with its enhanced health beneficial and desirable properties.

Phytochemical Analysis and Biological Evaluation of Hypericum linarioides Bosse: in Vitro and in Silico Studies

The aim of this study is to determine the phenolic composition of Hypericum linarioides Bosse, to assess its various biological properties, and finally to confirm the experimental data of enzyme inhibitions via docking studies. LC-MS/MS analysis performed over methanol extract (ME) revealed the presence of 27 phytochemicals. Antioxidant activity results indicated that ME had the strongest DPPH radical scavenging and ferric ion reducing capacity than water and hexane extracts. XTT assays revealed the strongest anti-proliferative activity of ME against MCF-7 and HT-29 cancer cells. Flow cytometry results also confirmed the notable apoptotic effect of ME on MCF-7 cells through mitochondria-dependent pathway. Enzyme activity results exhibited the most promising acetylcholinesterase and α-glycosidase inhibitory activities of the ME. Molecular docking calculations of α-glycosidase with the major compounds (1, 15, 17 and 19) indicated that 17 (isoquercitrin) had the best docking binding energy (À 10.92 kcal/mol) with appropriate binding interactions.

Nepeta is one of the largest genera of the Lamiaceae family. Nepeta species are commonly employed in traditional medicine for a variety of ailments, as well as food additives. In addition, they also come to the fore with their rich phytochemical content. In the present study, the quantitative phytochemical content of methanolic extracts and infusions prepared from the aerial parts of 14 Nepeta taxa collected from Turkey and their cytotoxic effects on two breast cancer cell lines, MCF-7 and MDA-MB-231, were investigated by using the MTT (3-(4,5-dimethylthiazol-2-yl))-2,5-diphenyltetrazolium-bromide) test. According to HPLC-PDA analysis, N. racemosa methanolic extract had the highest ursolic acid content with 165.9 mg/g extract. Total sterol, total iridoid, and total triterpenoid content were determined to be greatest in the methanolic extracts of N. meyeri, N. trichocalyx and N. phyllochlamys. The MTT experiment demonstrated that certain Nepeta species suppressed the growth of MCF-7 and MDA-MB-231 cells in a dose-dependent manner. Statistical analysis revealed a strong correlation between the cytotoxic effects of the extracts and their triterpene content. In conclusion, the data obtained from this study are important in terms of forming a basis for advanced anticancer activity studies on breast cancer with Nepeta sp.
