Antioxidant, Antibacterial Activities and Identification of Bioactive Compounds from Terminalia chebula Bark Extracts

Objective: The present study was undertaken to investigate the antioxidative activity of fruit pulp, seed and bark of T. bellerica. Methods: Methanol (70%) and water were used for the extraction, and the respective extracts were analyzed for total phenolic contents along with the antioxidant activities through different assays. Results: Amongst the tested extracts, methanolic extracts were found to be better than aqueous with the highest total phenolic contents (TPC). Methanolic fruit pulp showed lower IC50 for free radical (118.7 µg/ml), superoxide anion radical (77.65 µg/ml) and hydroxyl radical (73.76 µg/ml) scavenging activities. Methanolic fruit pulp also showed lower IC50 for lipid peroxidation (115.6 µg/ml) and ferric thiocyanate (184.98 µg/ml) assay than aqueous extracts in comparison to standard quercetin. Further, correlation between TPC and antioxidant studies revealed that phenolics are mainly responsible for antioxidant activity of T. bellerica. Conclusion: The current study suggests that the methanolic extract of T. bellerica could be a potential source of natural antioxidants for food and pharmaceutical companies.

Gereç ve Yöntemler: Kurutulmuş yaprak ve gövdelerden hazırlanan metanol ekstreleri üzerinde, vakum ve açık kolon kromatografısi metotları kullanılarak fraksiyonlama ve izolasyon çalışmaları yapılmıştır. Bileşiklerin yapıları, IR, 1D-ve 2D-NMR teknikleri kullanılarak ve literatürdeki verilerle ÖZ Objectives: Vitex grandifolia belongs to family Lamiaceae; it consists of flowering plants and it is also called the mint family. The Yoruba people of southwest Nigeria called it "Oriri" or "Efo oriri". This plant is classified as an underutilized vegetable and little is known about its phytochemistry or its biological evaluations. Materials and Methods: Methanol extracts of the dried leaves and stem of the plant were subjected to fractionation and isolation using vacuum layer and column chromatography methods. The structures of the compounds were elucidated using spectroscopic techniques including IR, 1D-, and 2D-NMR and by comparison with the data reported in the literature. They were evaluated in vitro for the inhibition of monoamine recombinant human MAO-A and-B and anti-inflammatory activities. Results: Three known flavonoids were isolated from the methanolic extract of the leaves of V. grandifolia for the first time to the best of our knowledge, i.e. isoorientin (1), orientin (2), and isovitexin (3). Most of the isolated compounds showed selective inhibition of monoamine oxidase B, inhibition of MAO-B by isoorientin (1) and orientin (2) were 9-fold more potent (IC 50 (µg/mL) of 11.08 and 11.04) compared to the inhibition of MAO-A (IC 50 (µg/mL) of ˃100), while clorgyline and deprenyl were used as positive standards. The isolated flavonoids displayed good activity against the NF-κb assay with IC 50 (µg/mL) of 8.9, 12, and 18. This study establishes a link between the structure and the biological activities on the basis of the different patterns of substitution, particularly the C2=C3 double bond and the position of glucose moiety. Conclusion: This study is the first to establish the phytochemistry of the polar part of V. grandifolia and the anti-inflammatory and neuroprotective role of these isolated compounds.

Plants have been used in traditional medicine for several thousand years since which may have been described to have curative value. Terminalia chebula (TC), Terminalia arjuna (TA) and Holarrhena antidysenterica (HA) are very important medicinal plants in the traditional Indian systems of Ayurveda and Siddha. In this work, fruit of Terminalia chebula, bark of Terminalia arjuna and Holarrhena antidysenterica were selected for experiments. 80% methanolic crude extracts of these plants were fractionated by diethyl ether and ethyl acetate. Diethyl ether fraction of 80% methanolic extract (DM) and ethyl acetate fraction of 80% methanolic extract (EM) of all the three plants were compared for their phytochemical, antioxidant and cytotoxicity to find out their therapeutic potential. The phenolic content was determined by Folin-Ciocalteu method and was found to be higher in Terminalia chebula extracts (>36 mg GAE/g)). The antioxidant activity was assayed through some in-vitro methods such as DPPH radical scavenging assay, phosphomolybdenum method and reducing power assay. Antioxidant activity was also found to be highest in Terminalia chebula extracts. Cytotoxicity of extracts was determined by the MTT assay using HeLa cell lines which showed that the increase in concentration of the extract increases the cell death. All the extracts showed moderate cytotoxicity with IC 50 values ranging from 147.91 to 1701 µg/ml. These findings showed that extracts from Terminalia chebula is a potential source of natural antioxidants and could be good alternatives to synthetic antioxidants in pharmaceutical industries.
