Infertility induced by anti-cancer treatments pose a major concern for cancer survivors. Doxorubicin (DXR) has been previously shown to exert toxic effects on the testicular germinal epithelium. Based upon the cardioprotective traits of dexrazoxane (DEX), we studied its potential effect in reducing DXR-induced testicular toxicity. Male mice were injected with 5 mg/kg DXR, 100 mg/kg DEX, combination of both or saline (control) and sacrificed either 1, 3 or 6 months later. Testes were excised and further processed. Glutathione and apoptosis assays were performed to determine oxidative stress. Immunohistochemistry and confocal microscopy were used to study the effects of the drugs on testicular histology and on spermatogonial reserve. DXR and the combined treatment induced a striking decline in testicular weight. DEX prevented DXR-induced oxidative stress, but enhanced DXR-induced apoptosis within the testes. Furthermore, the combined treatment depleted the spermatogonial reserve after 1 month, with impaired recovery at 3 and 6 months post-treatment. This resulted in compromised sperm parameters, testicular and epididymal weights as well as significantly reduced sperm motility, all of which were more severe than those observed in DXR-treated mice. The activity of DEX in the testis may differ from its activity in cardiomyocytes. Adding DEX to DXR exacerbates DXR-induced testicular toxicity. Reproduction (2015) 150 357-366

A direct interaction between p21 and p50 was confirmed by using ELISA and pull-down assays with purified proteins. The interaction sites between p50 and p21 were mapped by pull down assays with GST deletion mutants. Residues 127-193 constitute the primary interaction region on p50 to which p21 binds, while p50 binds to the C-terminal 26 residues of p21. A histone kinase activity was associated with the highly purified calf thymus pol δ and addition of purified recombinant p21 inhibited the kinase activity in a dose dependent manner. p50 is phosphorylated in vivo and can be phosphorylated by CDK2/cyclinA in vitro. In vivo evidence of p21 association with p50 was obtained by coimmunoprecipitation using MCF7 cells. It was also shown that the association of p21 with p50 and other components of the pol δ complex increased in MCF7 cells treated with adriamycin. Our results suggested that p50 might target or anchor p21 to pol δ complex upon certain DNA damage such as adriamycin treatment.

Turmeric is harvested with high moisture content and should be dried before the storage. It is observed that drying is quickest when the rhizomes are peeled and cut in small cylindrical pieces. In order to describe the process, normally a diffusive model is used, considering boundary condition of the first kind for the diffusion equation. This article uses analytical solutions considering boundaries conditions of the first (model 1) and third (model 2) kinds coupled to an optimizer to describe the drying process. It is shown that, for model 1, the fit of the analytical solution to the experimental data is biased, despite the good statistical indicators (chi-square χ 2 equal to 1.7095×10 −3 and coefficient of correlation R 2 of 0.9988). For model 2, the errors of the experimental points about the simulated curve can be considered randomly distributed, and the statistical indicators are much better than those obtained for model 1: χ 2 03.5596×10 −4 and R 2 00.9996.
