Antiproliferative and leishmanicidal effect of ajoene on various Leishmania species: ultrastructural study

Abstract: Ajoene [(E,Z)-4,5,9 trithiadodeca 1,6,11 triene 9-oxide], the major bioactive compound derived from garlic, shows a potent trypanolytic and antimicotic activity. In this paper we evaluate its effect on Leishmania mexicana(Lm:MHOM/VE/80/NR), L eishmania mexicana venezuelensis (Lmv: MHOM/VE/80/H16), L eishmania mexicana amazonensis (Lma: M112, IFLA/BR/67/PH8) and L eishmania donovani chagasi (Ldch: MHOM/BR/74/PP75). Ajoene showed a potent leishmanicidal activity in vitro against all species studied. Concentratio… Show more

“…Taken together, these results strongly support the potential of garlic extracts for the immunotherapy of Leishmania infection. This immunomodulatory activity of garlic extract may also be complemented by the direct inhibitory effect of some garlic compounds such as ajoene, which can significantly inhibit Leishmania growth in vitro [32].…”

“…Ajoene decreased intracellular L. mexicana infection by 40% at 10 µM, by 95% at 50 µM and by 100% at 100 µM while not affecting host macrophages. Ultrastructural studies showed a time-and dose-dependent morphological alteration of the mitochondrial membrane and nuclear envelope and the formation of large autophagic vacuoles and megasomes [160]. Ajoene (161) also induced cell lysis of T. cruzi epimastigotes at 100 µM.…”

“…Earlier, ajoene was found to be a potent inhibitor of Leishmania mexicana and Leishmania venezuelensis but at IC 50 2.0-4.5 μg/mL. [28] . Other studies reported that the susceptibility of amastigotes of Leishmania mexicana to pentostam and paromomycin was at concentrations of 4.1-21 and 24 μg/mL, respectively.…”

“…The results of current study revealed significant axenic anti‐amastigote and antipromastigote activity of hirudin with IC 50 values considerably lower than glucantime. Earlier, ajoene was found to be a potent inhibitor of Leishmania mexicana and Leishmania venezuelensis but at IC 50 2.0–4.5 μg/mL …”
