Antiproliferative effects of isopentenylated coumarins isolated from Phellolophium madagascariense Baker

Abstract: From the leaves of Phellolophium madagascariense Baker (Apiaceae), an endemic herb to Madagascar, three known coumarins (osthol, murraol and meranzin hydrate) have been isolated and identified. This is the first report of these compounds in this species. The structural elucidations were based on the analysis of physical and spectroscopic data. The anticancer activity of the three isolated compounds and of a synthetic sample of osthol was evaluated on L1210 mouse leukemia and on human prostatic cancer hormonose… Show more

“…Furthermore, the positive optical rotation value of 1 ([α] 25 +3.8 o ) was in good agreement with that of related (2'R)-hydroxy-coumarin derivatives. 15,16 The known compounds, osthenol (2), 17 (2'R)-7-hydroxy-8-(2',3'-dihydroxy-3'-methylbutyl)-2H-1-benzopyran-2-one (3), 18 columbianetin (4), 19 and seselin (5) 20 were also isolated. Their structures were identified by comparing their spectroscopic data with those in the literature.…”

“…Previous investigations have shown that osthole possesses various beneficial pharmacological activities including: anti-osteoporotic, anti-inflammatory, anti-allergic, anti-seizure, and anti-diabetic effects [12,13,14,15]. Moreover, accumulating evidence has shown that osthole exerts anti-cancer effects through inducing cell cycle arrest and apoptotic cell death [16,17,18]. Recent studies have also shown that osthole is able to inhibit the migration and invasion of cancer cells through suppression of MMP-2 and MMP-9 enzyme activities [19,20].…”
