Antisense‐mediated Depletion of Tomato Endoplasmic Reticulum Omega‐3 Fatty Acid Desaturase Enhances Thermal Tolerance

Abstract: An endoplasmic reticulum-localized tomato omega-3 fatty acid desaturase gene (LeFAD3) was isolated. The antisense tomato plants were obtained under the control of the cauliflower mosaic virus 35S promoter (35S-CaMV). Northern blot analysis confirmed that the expression of LeFAD3 was inhibited in the tomato genome. Levels of 18:3 decreased and correspondingly levels of 18:2 increased in total lipids of leaves and roots. After heat stress, the fresh weight of the aerial parts of antisense transgenic plants was h… Show more

“…GO Biological Process is helpful for functional classification of analyzed genes. Terms “response to abiotic or biotic stimulus” and “response to stress” are enriched (Figure 2C), which is consistent with the previous studies showing that wax and cutin are protective tissues for environmental stress and unsaturated FAs play important roles in various stresses (Samuels et al 2008; Upchurch 2008; Wang et al 2010). More detailed analysis of developmental process showed that the identified co‐expressed genes are involved in floral organs (ovule, stamen and carpel) and whorl development, shoot and leaf morphogenesis, cuticle development and post‐embryonic development (details in Table 2).…”

“…Many biotic and abiotic stresses (such as heat, cold, drought, salt, and wounding) can cause chloroplast damage in plants (Price et al 1989;Chang et al 2004;Gao et al 2007;Wang et al 2010;Zhao et al 2010;Gong et al 2014). Therefore, damage in chloroplasts can also cause similar responses to biotic and abiotic stresses.…”

“…The omega‐3 fatty acid desaturase gene was also found to be involved in high temperature tolerance in transgenic tomato, which had reduced activity of endoplasmic reticulum (Wang et al. ).…”
