“…Fusidik asit ABD'de lisanslı olmadığından, NCCLS önerileri içinde duyarlılık sınırına dair değer bulunmamaktadır (6). Ancak gerek İngiliz Antimikrobiyal Kemoterapi Derneği (BSAC), gerek Fransa Mikrobiyoloji Derneği, gerekse bu konuda standart oluş-turmak amacıyla yapılmış çalışmalar fusidik asit'in stafilokoklar için MİK değerlerini duyarlı, orta duyarlı ve dirençli olmak üzere sırasıyla; ≤0.125 mg/L, 0.5-1 mg/L ve ≥2 mg/L olarak bildirmektedir (8)(9)(10) 1962 yılında İngiltere'de yapılan iki çalışmada toplam 150 S.aureus suşunun 147'sinin (%98) fusidik asite duyarlı (MİK≤0.125mg/L), 3'ünün (%2) ise orta duyarlı oldukları (MİK=0.5mg/L) bildirilmiştir (15,16). Aradan geçen 40 yılı aşkın sürede fusidik asitin stafilokoklara karşı etkinliğini koruduğu görülmektedir.…”

Amaç: Bu çalışmada klinik örneklerden izole edilen ve enfeksiyon etkeni olarak kabul edilen 440 Staphylococcus aureus suşunun fusidik asite in-vitro duyarlılığı araştırılması amaçlandı. Gereç ve yöntem: "National Committee for Clinical Laboratory Standards" (NCCLS) önerileri doğ-rultusunda agar dilüsyon yöntemi ile minimal inhibitör konsantrasyon (MİK) değerleri belirlendi. MİK değeri ≥2 mg/L olan suşlar dirençli, ≤0.125mg/L olan suşlar duyarlı kabul edildi. Bulgular: Çalışmaya alınan 196 metisiline duyarlı S.aureus suşunda fusidik asite direnç saptanmazken, 244 metisiline dirençli S.aureus suşundan 2'si (%0.8) fusidik asite dirençli bulundu. Sonuç: Fusidik asit, metisiline dirençli ve duyarlı stafilokokların etken olduğu enfeksiyonlarda iyi bir tedavi seçeneği olarak karşımıza çıkmaktadır. Anahtar sözcükler: Staphylococcus aureus, fusidik asitAim: This study was undertaken to investigate the in vitro susceptibility of Staphylococcus aureus strains to fusidic acid. Materials and methods: A total of 440 strains isolated from various clinical specimens were included in the study. The susceptibility tests were performed by agar dilution method acccording to the NCCLS (National Committee for Clinical Laboratory Standards) criteria. Strains with MIC value of ≥2 mg/L were taken as resistant to fusidic acid. Results: None of the methicillin-susceptible S.aureus (MSSA) strains were found to be resistant while 2 (0.8%) of 244 methicillin-resistant S.aureus (MRSA) strains were resistant to fusidic acid. Conclusion: Fusidic acid can be considered as an alternative drug for the treatment of infections due to both methicillin susceptible and resistant S.aureus strains.

Moreover, fusidic acid penetrates the aqueous and vitreous humour of the non inflamed eye and has hence been used, in combination with other agents, for the treatment of postoperative intraocular infections (Chadwick and Jackson 1969;Williamson et al 1970;Kanski 1974).…”

Fifty years of drug discovery from fungi

For the past 50 years, fungal secondary metabolites have revolutionized medicine yielding blockbuster drugs and drug leads of enormous therapeutic and agricultural potential. Since the discovery of penicillin, the first β-lactam antibiotic, fungi provided modern medicine with important antibiotics for curing life threatening infectious diseases. A new era in immunopharmacology and organ transplantation began with the discovery of cyclosporine. Other important drugs or products for agriculture derived from or inspired by natural products from fungi include statins, echinocandins and strobilurins. Moreover, fungal biotransformation of steroids for the industrial production of steroidal hormones represents one of the key successes in biotechnology. Given that estimations of fungal biodiversity exceed by far the number of already identified species, chances to find hitherto unidentified fungal species and novel bioactive fungal products are still high. Thus, further compounds with medicinal or agricultural potential from less investigated fungal taxa can be expected in the years to come.
