Antiuveitis and Inhibition of Fibroblast-Like Corneal and Conjunctival Cell Cultures by Interleukin-1 Blockers, CK 125, CK 126 and CK 128

Abstract: Three new interleukin-1 (IL-1) blockers, CK 125, CK 126 and CK 128, were studied for their effects on IL-1-induced uveitis in rat eyes. They were more potent (at 3–10 mg/kg t.i.d.) than prednisolone (20 mg/kg t.i.d.) in effectively inhibiting posterior uveitis. They were also found to inhibit fibroblast-like corneal cells at 10–300 µg/ml concentrations and conjunctival cells at 1–30 µg/ml levels. The incorporation of leucine into corneal and conjunctival cells was either stimulated or unaffected by CK 126, ind… Show more

“…The recent discovery of synthetic IL-1 blockers which produce potent anti-inflammatory actions with few sideeffects, if any, is most exciting and warrants further studies [1,[4][5][6]. It has been found in this study that CK130 is the most potent anti-inflammatory agent of approximately 150 compounds investigated [1,[4][5][6][7][8][9][10][11].…”

“…Because of their anti-inflammatory actions, IL-1 blockers can also be used as inhibitors of fibroblast-like corneal and conjunctival cell proliferation [7][8][9][10][11][12]. If effective, these compounds could also be used to improve the success rate and functional period of cannula created after filtration surgery (trabeculectomy) in closeangle and low-tension glaucoma treatments [13,14].…”

“…Numerous agents, including natural products and synthetic compounds, have been found to block IL-la-induced uveitis (23)(24)(25)(26)(27)(28)(29)(30)(31). These agents include tetrandrine (23), chalcone derivatives (24,25), osthole (26), and synthetic organic CK-compounds (27)(28)(29)(30)(31).…”
