(1) Background: Largemouth bass virus (LMBV) is a major viral pathogen in largemouth bass (Micropterus salmoides) aquaculture that often causes high mortality and heavy economic losses, thus developing treatments to combat this pathogen is of great commercial importance. Green tea is a well-known medicinal plant that contains active ingredients with antiviral, antibacterial, and other biological activities. The goals of this study were to explore the effect and mechanism of green tea source compounds on LMBV and provide data to serve as the basis for the screening of targeted drugs in the future. In this study, we evaluated the effects of the main component of green tea, epigallocatechin-3-gallate (EGCG), against LMBV infection. (2) Methods: The safe working concentration of EGCG was identified by cell viability detection and light microscopy. The antiviral activity and mechanism of action of EGCG against LMBV infection were evaluated with light microscopy, an aptamer 6-carboxy-fluorescein-based fluorescent molecular probe, and reverse transcription quantitative PCR. (3) Results: The safe working concentration of EGCG was ≤10 μg/mL. EGCG showed significant anti-LMBV infection activity in a concentration-dependent manner, and it also destroyed the structure of virus particles. EGCG impacted the binding of virus particles to cell receptors and virus invasion into the host cells. Inhibitory effects of EGCG on LMBV particles, LMBV binding to the host-cell membrane, and LMBV invasion were 84.89%, 98.99%, and 95.23%, respectively. Meanwhile, the effects of EGCG subsequently were verified in vivo. The fatality rate of the LMBV + EGCG group was significantly lower than that of the LMBV group. (4) Conclusions: Our results suggest that EGCG has effective antiviral properties against LMBV and may be a candidate for the effective treatment and control of LMBV infections in largemouth bass aquaculture.

Regulation of antibiotic use in aquaculture calls for the emergence of more sustainable alternative treatments. Tea polyphenols (GTE), particularly epigallocatechin gallate (EGCG), have various biological activities. However, tea polyphenols are susceptible to degradation. In this work, EGCG and GTE were encapsulated in zein nanoparticles (ZNP) stabilized with alginate (ALG) and chitosan (CS) to reduce the degradation effect. ALG-coated ZNP and ALG/CS-coated ZNP encapsulating EGCG or GTE were obtained with a hydrodynamic size of less than 300 nm, an absolute ζ-potential value >30 mV, and an encapsulation efficiency greater than 75%. The antioxidant capacity of the encapsulated substances, although lower than that of the free ones, maintained high levels. On the other hand, the evaluation of antimicrobial activity showed greater efficiency in terms of growth inhibition for ALG/CS-ZNP formulations, with average overall values of around 60%, reaching an inhibition of more than 90% for Photobacterium damselae. These results support encapsulation as a good strategy for tea polyphenols, as it allows maintaining significant levels of antioxidant activity and increasing the potential for antimicrobial activity, in addition to increasing protection against sources of degradation.

This paper attempts to analyze and assess Yongchuan Xiuya tea quality quickly, accurately, and digitally. The sensory evaluation method was first used to assess Yongchuan Xiuya tea quality, and then near infrared spectroscopy (NIRS) was obtained, and standard methods were applied to the testing of the chemical components. Next, principal component analysis (PCA) and the correlation coefficient method were used to comprehensively screen out the representative components. Finally, NIRS combined with partial least squares regression (PLSR) and back propagation artificial neural network (BP-ANN) methods were applied to build quality evaluation models for Yongchuan Xiuya tea, respectively, and external samples were employed to examine the practical application results of the best model. The cumulative variance contribution rate of the first three principal components of the ingredients in tea was 97.73%. Seven components closely related to tea quality were screened out, namely, amino acids, total catechin, epigallocatechin gallate (EGCG), tea polyphenols, water extracts, epicatechin gallate (ECG), and epigallocatechin (EGC) (p < 0.01). Between the two models established to predict the tea quality, the model built by the PLS method had the better results, whose coefficient of determination of prediction (Rp2) and root mean square error of prediction (RMSEP) were 0.7955 and 1.2263, respectively, and the best results were obtained by the nonlinear BP-ANN model, whose Rp2 and RMSEP were 0.9315 and 0.6787, respectively. The 10 external Yongchuan Xiuya samples were employed to test the best BP-ANN model, and the results of R2 and RMSEP were 0.9579 and 0.6086, respectively, meaning that the model has good robustness. Therefore, the model established by NIRS combined with the BP-ANN method can be used to assess Yongchuan Xiuya tea quality rapidly, accurately, and digitally, and it can also provide new ideas and methods for evaluating the quality of other teas.
