Antiviral therapy in shrimp through plant virus VLP containing VP28 dsRNA against WSSV

Abstract: The white spot syndrome virus (WSSV), currently affecting cultured shrimp, causes substantial economic losses to the worldwide shrimp industry. An antiviral therapy using double-stranded RNA interference (dsRNAi) by intramuscular injection (IM) has proven the most effective shrimp protection against WSSV. However, IM treatment is still not viable for shrimp farms. The challenge is to develop an efficient oral delivery system that manages to avoid the degradation of antiviral RNA molecules. The present work dem… Show more

“…The virus infection titers (SID 50 mL −1 ) and mortality (LD 50 mL −1 ) can subsequently be calculated using the method of Reed and Muench [178]. Since this procedure was described in shrimp [51,53,105], it has been used in multiple studies to determine the infective titers of WSSV stocks so that a known dose of infectious virus could be administered to a group of susceptible individuals [10,76,102,115,121,179]. Through this method, Rahman et al [115], for instance, managed to demonstrate that virulence differences exist between WSSV geographic isolates.…”

“…For instance, vp28-siRNAs injected along with WSSV in M. japonicus were shown to inhibit WSSV replication [96,353,354]. Moreover, the use of vp28 and vp37-dsRNA effectively eliminated WSSV virions from M. japonicus [355] and L. vannamei [179,250] challenged with WSSV [352]. Most studies have applied intramuscular RNAi administration [179,250,[356][357][358][359][360][361][362][363][364][365].…”

“…These specific dsRNA fragments, known as small interfering RNAs (siRNAs), confer sequence specificity in subsequent steps that lead to the degradation of target viral mRNA inhibiting viral replication [ 63 ]. In shrimp, the use of vp28 and vp37 siRNA effectively eliminates WSSV virions from M. japonicus [ 64 , 65 ] and Litopenaeus vannamei (Boone) [ 66 , 67 ] shrimp, challenged with WSSV, demonstrating the efficiency of siRNA-based RNAi as a technique to combat viral infections in shrimp. YHV-specific dsRNA-injected shrimp targeting the RdRp and rr2 genes do not exhibit yellowhead disease after the YHV challenge [ 68 , 69 , 70 ].…”

“…Numerous research investigations have employed sequence-specific dsRNA to impede the replication of viruses such as TSV, IHHNV, YHV, WSSV, LSNV, and GAV in shrimp [ 71 , 87 , 88 , 90 , 103 ]. These studies involve the targeted suppression of particular genes such as Vp28 [ 67 ], Vp37 [ 65 ], rr2 [ 70 ], V9 [ 97 ], rab7 [ 80 ], a caspase-3 protein [ 131 ], and others ( Table 2 ).…”

“…The proteins of most viruses assemble in an icosahedral or helical symmetry around their genome. This is a self-assembly process that has also been observed only with the protein subunits without their genome [ 21 ], and there are more complex assemblies requiring additional viral (or host) factors [ 22 , 23 , 24 ]. An icosahedron is defined by 20 triangular faces and 12 vertices related by two-, three-, and five-fold axes of rotational symmetry [ 25 ].…”
