The increasing prevalence of anxiety and stress-related disorders has made it a leading contributor to the global health burden. The present treatment options have severe side effects and show remission on discontinuation of the medication. Hence, there is an urgent need to explore safer alternative treatments for long-term usage with minimum toxicity. The medicinal plant Brahmi (Bacopa monnieri) has been used in Indian traditional medicine as a neural tonic for centuries. The present study aimed to study the toxicity and anxiolytic activity of Brahmi using the zebrafish model system. The toxicity assays determined the minimum effective concentration of Brahmi to be 0.01%. In addition, behavioral assays such as thigmotaxis and scototaxis and endocrine assays such as the measurement of cortisol levels in stressed zebrafish larvae were performed. Zebrafish embryos exposed to 0.2% Brahmi up to seven days post fertilization (dpf) did not show any developmental toxicity. Behavioral and endocrine assays were performed on 5dpf zebrafish larvae treated with 0.01% Brahmi extract. Our studies show that Brahmi significantly reduced thigmotaxis (wall hugging) and scototaxis behavior in zebrafish larvae exposed to osmotic stress as compared to untreated stressed larvae. Stress activates the hypothalamic-pituitary-interrenal axis and stimulates the release of cortisol in zebrafish larvae. Whole body cortisol assay has shown that Brahmi significantly reduced the stress-induced release of cortisol in zebrafish larvae. Our studies report that Brahmi mitigates the stress response in zebrafish larvae and has minimum toxicity. This suggests that Brahmi may be a safe option for long term management of stress.

Throughout history, complementary and alternative therapies have been widely utilised. In recent years, there has been a surge in interest in the usage of herbal treatments all around the world. Various natural chemicals, such as those produced from plants, have been investigated as potential therapies for a myriad of ailments. The essence of this review was to methodically describe everything we know about Bacopa monnieri (L.) Pennell, a mysterious holistic Vedic herb belonging to the Plantaginaceae family, a well-known nootropic and effective memory enhancer, which has recently emerged as one of the most important medical herbs, widely used therapeutically in the Orient and growing in popularity around the world. Literature was gathered from sources such as Scopus, PubMed, Google Scholar, and ScienceDirect, and reviewed using the Prisma quality metacritic paradigm. It is now plainly obvious that current therapies fall short of meeting the demands of the vast majority of individuals with health problems, and traditional medicines are gaining appeal as a result of their reduced toxicity. Bacopa is a traditional herb used in Ayurvedic medicine to treat brain and nerve weariness, as well as in Siddha medicine to treat impaired memory. It's also used to cure brain and nerve exhaustion in Unani medicine. We improved Brahmi micropropagation and secondary metabolite biosynthesis by compiling pharmacobotanical and pharmacognostical descriptions, as well as ethnoarchaeological data and nanotechnology domination. This critique also highlights our contemporary information of pharmacological activity, preclinical and clinical investigations, significant bioactives, reported mechanisms of action, clinical effectiveness, safety, and the potential for herb-drug interactions. At the same time, the current incarnation of research at the plant is reviewed, as well as future research possibilities. Brahmi offers a lot of potential for treating a range of illnesses, including neuro-pharmacological, depression, inflammation, hepatoprotective, antidiabetic, and others. According to the presumptions of this review, further clinical trials and research are needed. While the impact of Brahmi as an anxiolytic and antidepressant has to be explored further, its potential as an anti-epileptic therapy and a treatment for antiepileptic drugs side effects is also being researched. Furthermore, Brahmi's antioxidant ability may explain, at least in part, the antistress, immunomodulatory, cognition-facilitating, anti-inflammatory, and anti-aging benefits documented in experimental animals and clinical circumstances, necessitating further study into its other therapeutic characteristics.

Aims: The study determined and compared the herbal extraction yields using water, ethanol and hydromethanol solvent and the solvent extracting the highest antidiabetic constituents. Place: The study took place in the Department of Chemistry (Organic Laboratory), Federal University of Technology Owerri, Nigeria. Methodology: The antidiabetic contents of Moringa oleifera (Moringa) and Vernonia amygdalina (bitter leaf) were extracted by soaking using water, ethanol and hydromethanol (1:1) as solvents. The phytochemicals analysis was done both qualitatively and quantitatively (using Spectrophotometer (UV-V15)). Data collected were statistically analysed using SPSS version 10 tools. Results: The crude ethanolic extraction was found to give the highest extract yield of 46.06% and 38.91% from Moringa and bitter leaf, respectively. There were more phytochemicals obtained from Moringa (28+) than from Vernonia crude extracts (21+). The antidiabetic phytochemicals identified in both plants included Steroids, Phenols, Cardiac glycosides and Terpenoids. Ethanol extracted the Glycosides, Terpenoids and Phenols in relative abundance.. Hydromethanol solvent extraction yielded the highest concentrations of Steroids from Moringa (59.87mg/100g) and bitter leaves (75.43mg/100g) as well as highest extraction of Cardiac glycosides from both plants. Water extracted the highest concentrations of Phenols from both Moringa (0.32mg/g); bitter leaf (0.25mg/g) and Terpenoids from Moringa. Conclusion: This study suggests that the choice of solvent (s) for phytochemical extraction (s) should consider factors such as the plant material (s) and the phytochemical (s) involved. So, Water > Hydromethanol > ethanol could be used for extracting phytochemicals for diabetes therapy.
