Anxiolytic-Like and Antidepressant-Like Activities of MCL0129 (1-[(S)-2-(4-Fluorophenyl)-2-(4-isopropylpiperadin-1-yl)ethyl]-4-[4-(2-methoxynaphthalen-1-yl)butyl]piperazine), a Novel and Potent Nonpeptide Antagonist of the Melanocortin-4 Receptor

Abstract: ABSTRACT]-␣-melanocyte-stimulating hormone (␣-MSH) binding to MC4 receptor with a K i value of 7.9 nM, without showing affinity for MC1 and MC3 receptors. MCL0129 at 1 M had no apparent affinity for other receptors, transporters, and ion channels related to anxiety and depression except for a moderate affinity for the 1 receptor, serotonin transporter, and ␣ 1 -adrenoceptor, which means that MCL0129 is selective for the MC4 receptor. MCL0129 attenuated the ␣-MSHincreased cAMP formation in COS-1 cells expressin… Show more

“…7) Recently, a nonpeptide MC4 receptor antagonist was reported to exhibit antidepressant-like activity in rodent models. 8) Evaluating the interaction of MC4R with a compound (an antagonist or an inverse agonist) requires the generation of constitutively activated MC4R. Some receptors were constitutively activated by introducing mutagenesis at the DRY conserved sequence, which is located at the amino terminus of the second intracellular loop of the 7TM receptors.…”

“…The MC4 receptor has been reported to play important roles in regulation of the activity of the HPA axis (20,21). Moreover, a MC4-receptor antagonist has revealed the involvement of this receptor in emotional states such as anxiety and depression (22). On the other hand, an MC4-receptor agonist depressed the firing rate of the dorsal raphe nucleus of serotonergic neurons (23).…”

“…[163][164][165][166][167][168][169][170][171][172] This has been explained in part by the fact that dopaminergic fibres from the VTAproject to the NAcc through the lateral hypothalamus. Studies of feeding behaviour have especially provided new insights into the interaction between hypothalamus and VTA-NAcc.…”
