APH-1, a POU homeobox gene expressed in the salt gland of the crustacean Artemia franciscana

Abstract: We characterized the first POU-homeoprotein in a crustacean (designated APH-1 for Artemia POU-Homeoprotein, EMBL Y15070). The amino acid sequence of the APH-1 POU-domain is identical, except for two residues, to that of the two class III POU proteins Cf1-a (Drosophila) and POU-M1 (Bombyx mori). Southern blot analysis suggests that crustaceans have only one class III POU gene. RT-PCR and whole-mount in situ hybridization show that APH-1 mRNA is present in larvae specifically in the salt gland, an organ which is… Show more

“…Tracheal cells and spiracles on T2 and T3 arise from locations lateral to the leg bases (23) possibly homologous with the association between gills and appendages in crustaceans, a hypothesis strengthened by the finding that homologs of tracheal inducer genes (trh, vvl) are specifically expressed in the gills of crustaceans (24,41). Notably, those genes were absent from the small T1 ectopic wings of Oncopeltus.…”

“…An antibody against the Vvl protein of Artemia was generated by injecting rabbits with a His-tagged fragment of the Vvl protein [amino acids 241 to 386 of the previously published sequence (Chavez et al, 1999)]. The serum was then affinity purified on a nickel column.…”

“…In the case of Artemia vvl, we cloned a fragment that corresponds to the APH-1 gene previously reported by Chavez et al (Chavez et al, 1999) and generated an antibody for immunochemical staining in developing Artemia larvae. We observe that Artemia Vvl is initially absent from early limb buds; it becomes weakly and uniformly expressed while the limb is developing its characteristic branching morphology, and becomes strongly upregulated in one of the epipods as its cells begin to differentiate (Fig.…”

“…In situ hybridization as a means to detect the RNA expression in cells and tissues has been widely described for D. rerio, Artemia, Drosophila and other species (Chavez et al, 1999;Mitchell and Crews, 2002;Thisse and Thisse, 2007), but studies on small cladocerans are much less common. In this study, we analyzed the spatial expression sites of DapcaDsx2 to ensure the integrity of D. carinata individuals by using digoxigenin-labeled RNA probes for the first time.…”

“…The whole-mount in situ hybridization technique employed the methods used by Thisse et al (Chavez et al, 1999;Mitchell and Crews, 2002;Thisse and Thisse, 2007), with changes based on the characteristics of D. carinata. Individual specimens were digested with proteinase K for about 15 min.…”
