Apnea stimulates the adaptive response to oxidative stress in elephant seal pups

Abstract: SUMMARYExtended breath-hold (apnea) bouts are routine during diving and sleeping in seals. These apneas result in oxygen store depletion and blood flow redistribution towards obligatory oxygen-dependent tissues, exposing seals to critical levels of ischemia and hypoxemia. The subsequent reperfusion/reoxygenation has the potential to increase oxidant production and thus oxidative stress. The contributions of extended apnea to oxidative stress in adapted mammals are not well defined. To address the hypothesis th… Show more

“…Adaptations that potentially decrease such risks in marine mammals include hypoxic pulmonary vasodilatation and elevations in both antioxidant concentrations and antioxidant enzyme activities (Zenteno-Savín et al, 2002;Olson et al, 2010;Tift et al, 2011;Vázquez-Medina et al, 2011). We propose that the levels of heme degradation, and associated by-products such as endogenous CO found in elephant seals, may also contribute to the protection against these conditions.…”

“…The increase in the number and duration of sleep apneas is associated with an increase in antioxidants (Vázquez-Medina et al, 2010;Vázquez-Medina et al, 2011b) over the course of the post-weaning fast. Furthermore, it was recently demonstrated that plasma XO activity, HX and xanthine content increase in elephant seal pups in response to rest-and voluntary submersion-associated apneas (Vázquez-Medina et al, 2011c). Therefore, fasting and sleep apnea may stimulate ATP degradation, increasing intracellular and circulating oxidant production, potentially modulated by increased purine salvage/recycling pathways (Elsner et al, 1998;ZentenoSavín et al, 2002).…”

“…Biopsies were rinsed with cold, sterile saline, placed in cryogenic vials, immediately frozen by immersion in liquid nitrogen, and stored at −80°C until later analyses. Frozen tissue samples were homogenized in 500 μl of hypotonic buffer containing a protease and phosphatase inhibitor cocktail (Sigma-Aldrich, St Louis, MO, USA) as previously described (Vázquez-Medina et al, 2011;Viscarra et al, 2011a;Viscarra et al, 2011b;Vázquez-Medina et al, 2012;Viscarra et al, 2012). Nuclear and cytosolic extractions were performed using the Nuclear and Cytoplasmic Extraction Reagents Kit (Thermo Fisher Scientific, Waltham, MA, USA).…”

“…Because our hypothesis focused on the changes in the deiodinases and the TH receptor, these proteins were given priority. Protein expression was quantified by standard western blot as previously described (Vázquez-Medina et al, 2011;Viscarra et al, 2011a;Viscarra et al, 2011b;Vázquez-Medina et al, 2012;Soñanez-Organis et al, 2012;Viscarra et al, 2012). The primary antibodies for DI1 and DI2, THrβ1, TATA binding protein, histone H3 and β-actin (Santa Cruz Biotechnology, Santa Cruz, CA) were diluted 1:500 to 1:5000.…”
