Apocynin Is Not an Inhibitor of Vascular NADPH Oxidases but an Antioxidant

Abstract: Abstract-A large body of literature suggest that vascular reduced nicotinamide-adenine dinucleotide phosphate (NADPH) oxidases are important sources of reactive oxygen species. Many studies, however, relied on data obtained with the inhibitor apocynin (4Ј-hydroxy-3Јmethoxyacetophenone). Because the mode of action of apocynin, however, is elusive, we determined its mechanism of inhibition on vascular NADPH oxidases. In HEK293 cells overexpressing NADPH oxidase isoforms (Nox1, Nox2, or Nox4), apocynin failed t… Show more

“…We reported similar observations related to coronary vascular dysfunction in ZO rats (Katakam et al, 2005). It has been proposed that in endothelial cells and vascular smooth muscle cells, apocynin reduces ROS not by inhibiting NAPDH oxidase but possibly by scavenging ROS (Heumuller et al, 2008). However, it has been argued that whether apocynin inhibits NADPH oxidase in nonphagocytic cells without myeloperoxidase depends on the cell type studied and the ability of vascular wall to activate apocynin (Touyz, 2008).…”

“…2c). As apocynin is known to act as a radical scavenger [35] and the assembly of p47 phox and p67 phox subunits is required for NOX enzymatic activity [36], N-ASCs endogenously depleted of p47 phox (ESM Fig. 4a) were evaluated for ROS Fig.…”

“…The present results thus confirm that the function of the vascular smooth muscle cells is not significantly affected by the endothelial regeneration process. 30,31 Incubation for 30 min with apocynin [10 −4 M; antioxidant and inhibitor of NADPH oxidase 35,36 ] or BMS309403 [3 × 10 −6 M; A-FABP inhibitor 23,37 ] had no significant effect on the responses, in terms of EC 50 or maximal response, of porcine coronary arteries with either native or regenerated endothelium ( Figure 2; Table 1) to serotonin, bradykinin, detaNONOate, and isoproterenol. Therefore, any effect (shown below) observed with these inhibitors after chronic treatment cannot be attributed to acute pharmacological actions per se of these agents.…”
