Apolipoprotein C3 facilitates internalization of cationic lipid nanoparticles into bone marrow-derived mouse mast cells

Abstract: Mast cells (MCs), are hematopoetically-derived secretory immune cells that release preformed as well as de novo synthesized inflammatory mediators in response to activation by several stimuli. Based on their role in inflammatory responses, particularly in the lung and skin, MCs provide an effective target for anti-inflammatory therapeutic strategies. Drug-delivery of lipophilic payloads to MCs can be challenging due to their functionally distinct intracellular structures. In the present study, pH-sensitive cat… Show more

“…Mast cells are highly granular/vesicular cells 5759 , it would be interesting to gain a better understanding of how they could incorporate such quantities of LNP. These findings are in line with previous reports that LNP could efficiently transfect in vitro -cultured mast cells 60,61 . While DC/macrophages are professional APCs that can initiate the activation of the adaptive immune response, less is known about the capacity of mast cells to present antigens to T cells in the context of vaccination.…”

“…[58] Conversely, in a different study, of all apolipoproteins tested, only APOC3 facilitated internalization of lipid-based nanoparticles into mast cells, potentially through APOE receptor 2 binding. [59] Curiously, since astrocytes and microglia are derived from the same glial precursors, it could be presumed that internalization occurs through a similar mechanism, despite apolipoproteins not showing correlation with uptake in astrocytes here. However, the field of nanomedicine is rapidly evolving, and novel roles for various apolipoproteins, including APOC3, in promoting cellular uptake might emerge with further research.…”
