“…The present study, for the first time, demonstrates that IFI27 protein stability was prolonged by EGF treatment in keratinocytes. By the mechanism of stabilizing proteins to enhance their activities, two other growth‐related proteins, NGFI‐B and EGR1 have also been found to be increased and stabilized by EGF . It is still not clear how EGF increase protein stability; however, our data provide evidence to support the role of IFI27 in pathogenesis of psoriasis, specially in hyperproliferation of keratinocytes.…”

“…Epidermal growth factor (EGF), an activating signal for ERK2, causes phosphorylation of Nur77 at threonine 142 [36], a phosphorylation site that stabilizes Nur77 [37], consistent with the phosphorylation-mediated stability of other nuclear receptors. Interestingly, EGF enhances the interaction of Nur77 with prolyl isomerase Pin1, and the isomerization of phosphorylated Nur77 requires ERK2- (and JNK1)-mediated phosphorylation [38].…”

“…Western analysis showed that NGFI‐B protein was present in cultures of rat cerebellar granule neurons at DIV 4/5 in concordance with former studies (Mathisen et al, 2011). The applied antibody targeting NGFI‐B had previously been validated (Mathisen et al, 2011; Strom and Paulsen, 2012) and a positive control was included to identify correct band size (results not included). Three siRNA sequences were tested, and the sequence specified in Section 2 was found to be effective.…”

“…NGFI-B is rapidly induced after stimuli (Watson and Milbrandt, 1989) and has a short half-life (Strom and Paulsen, 2012) which makes it a suitable candidate to target with siRNA. To provide reliable results this depends on high transfection efficiency.…”
