Apoptotic Activity and bcl-2 Immunoreactivity in Meningiomas

Abstract: We retrospectively evaluated 90 meningiomas for bcl-2 expression, apoptosis counts (per 10 high-power fields [HPF]), MIB-1 labeling indices (LI), and mitosis counts (per 10 HPF). Characteristics were as follows: 37 low-grade (benign) meningiomas: mean apoptosis count, 1.2; MIB-1 LI, 1.0; mitosis count, 0.1; and bcl-2 positivity, 40%; 29 atypical meningiomas: apoptosis count, 3.3; MIB-1 LI, 5.5; mitosis count, 2.2; and bcl-2 positivity, 62%; 24 malignant meningiomas: apoptosis count, 6.5; MIB-1 LI, 12.0; mitosi… Show more

“…Although the classical form of necrosis is a major histological factor influencing grading and prognosis in meningiomas [1][2][3], the occurrence and significance of apoptosis has been the subject of limited reports [4][5][6][7].…”

“…22 This is supported by the greater than 70% occurrence of protooncogene messenger RNA expression for c-myc and c-fos in meningiomas. Mutation in the tumor suppressor gene TP53 has been considered a reliable marker for malignant transformation of meningioma, 87 and the bcl-2 protooncogene is also correlated with higher-grade 1 Furthermore, expression of the ROS1 oncogene for tyrosine receptor kinase is common in meningiomas, which indicates that it may play a role in the origin of these tumors. 96 Although no single oncogene has been directly implicated in meningioma development, it is possible that either one or multiple oncogenes mentioned earlier will contribute to this process.…”

“…Interestingly, anaplastic meningiomas have been reported to exhibit a high apoptosis index, 4,5 and apoptosis is thought to be associated with atypical or malignant changes in these tumors. 30 The increased level of apoptosis in atypical and anaplastic meningiomas is associated with increased expression of the pro-apoptotic protein Bax and decreased expression of the anti-apoptotic protein Bcl2 in these cells.…”

“…3 Atypical and malignant meningiomas are histologically characterized by frequent mitoses, increased cellularity, high nuclear to cytoplasmic ratio, uninterrupted, patternless or sheet-like growth, and foci of necrosis. 2 In addition, anaplastic meningiomas exhibit an increased apoptosis index 4,5 and express higher levels of p53 and Bax and lower levels of Bcl2. 6,7 Fas-APO1 (CD95), is a 48-kd transmembrane cysteine-rich glycoprotein that belongs to the tumor necrosis factor receptor superfamily.…”
