Cordycepin is an active component extracted from Traditional Chinese medical herb Cordyceps militaris. Many reports demonstrated that cordycepin harbors antitumor activity in a broad spectrum of cancer types. In this study the actions and the underneath molecular mechanisms of cordycepin were investigated in renal cell carcinoma Caki-1 cell line. Results showed that cordycepin induced apoptotic cell death and inhibited cell migration in Caki-1 cells. Quantitative real-time PCR results and western blot analyses indicated cordycepin dose-dependently decreased microRNA-21 expression and Akt phosphorylation levels in Caki-1 cells, but increased PTEN phosphatase levels. Block of cordycepin-induced microRNA-21 decrease or PTEN increase in Caki-1 cells by transfection of microRNA-21 mimic or PTEN siRNA significantly attenuated cordycepin-induced cell death and inhibition of cell migration. Taken together, findings in present study suggested that cordycepin induced apoptotic cell death in renal cell carcinoma through regulation of microRNA-21 and PTEN phosphatase. Furthermore, present study also firstly illustrated that cordycepin inhibited cell migration of renal cell carcinoma, which also involved micro-RNA-21 and PTEN phosphatase.Renal cell carcinoma (RCC) is the most lethal urological cancer, accounting for 87% of all renal malignancies and 2-3% of all cancers (4). In the past two decades, the global incidence of RCC has increased by 2% per year (4). Worldwide, the agestandardized incidence and mortality of RCC were 6 per 100,000 and 2.1 per 100,000 respectively (43). Despite advance in diagnostic techniques, up to 20-30% of patients with RCC present with metastasis (4). The prognosis of RCC has historically been poor, with 5-year survival rate of 8% among patients with metastatic RCC (9). Although recent progress about understanding of the pathogenesis of RCC has led to novel immune-based and targeted treatments for this chemoresistant cancer, no one drug obtains overall response rate of 100% and durable complete responses (4, 9). Development of neo-adjuvant therapy is still in a demand at present. The aim of the present study was to examine the apoptotic effect of cordycepin (COR) on human THP-1 acute monocytic leukemia cells. THP-1 cells were exposed to different concentrations of COR for 24, 48, 72 or 96 h. The cell viability and apoptotic rate were analyzed. The gene expression of Akt1, Akt2, Akt3, B-cell lymphoma 2 (Bcl-2) and Bcl-2-associated X protein (Bax) were assessed by reverse-transcription quantitative PCR. Western blot analysis was used to detect the protein levels of phosphorylated (p)-Akt, p-extracellular signal-regulated kinase (ERK) and cleaved caspase-3. It was found that the viability of THP-1 cells was inhibited by COR in a dose- and time-dependent manner. After treatment with 200 µM COR for 24 h, the percentage of apoptotic cells was significantly increased. COR also downregulated the levels of Bcl-2, Akt1, Akt2 and Akt3, and elevated the expression of Bax. The protein levels of p-Akt and p-ERK were suppressed and cleaved caspase-3 was increased after treatment of COR. In conclusion, COR was found to induce apoptosis of THP-1 acute monocytic leukemia cells through downregulation of ERK/Akt signaling.

According to the reports from Ministry of Health in each country, the average life spans were expanded over the past century. This trend is expected to be extended further in the future and at the same time continuous efforts should be made. That indicates that the quantitative objective, as one of the objectives people have aimed at since ancient times to attain the perpetual youth and longevity, are almost achieved in this way. However, as the next objective, it is necessary for us to be concerned about improving the more advanced quality of life.In an effort of improving the quality of life, other than the western medicine, we have attempted to bring many traditional medical practices, including the Oriental medicine, from various parts of the world into the medical field as alternative medicine. Judging both positive and negative aspects by the evaluation standard of the Western medicine, we try to obtain numerical values of QOL, and after that, set a numerical target for improvement of QOL. Some methods of alternative medicine taken in this review are acupuncture moxibustion, Chinese medicine and other traditional medicine. Above all, WHO suggests selecting the suzerain nation of acupuncture moxibustion, which has been developed in the Oriental countries and realized as a specialized cure, by raising good & bad points and ascertaining which nation advocating the best modeling method could be. At the moment for evaluating CAM, for example, what kind of methods is/are suitable for evaluating each CAM. We have been trying to propose the peripheral leukocyte is one of the best ways for evaluating CAM. Our trials are hot spring hydro therapy, acupuncture and moxibution, light exercise and undle heating. In these evaluations, we found the common results from leukocyte effects which exhibited strong correlation for the regulation. The contents are the result from the data showing the much in number tend to downly regulate, on the other hand lower number one is up-regulated. This kind of regulation showed within a 24 hrs, for the leukocyte subsets, granulocyte and lymphocyte are changed under circadian rhythm. So for purpose of evaluating leukocyte deviation, we set the point for evaluation at the same-time zone from the first set of evaluation. Under these condition, we got a same results reproductively that the whole number of leukocyte, and its subset, granulocyte and lymphocyte also regulated within a 24 hrs. This kind of phenomenon is the case that we confirm a lot of kind of CAM and repetition. However, the life span of leukocyte are at lease 3-4 days and no such a drastic apoptosis was induced such a stimulation. For the purpose of scientific explanation, we proposed that the emotional hormone are concern this change of leukocytes population. For the results of this hypothesis, there were reasonable change were seen in the peripheral blood about emotional hormone, adrenaline and dopamine.
