APPARENT LIGHT REQUIREMENT FOR ACTIVATION OF PHOTOSYNTHESIS UPON REHYDRATION OF DESICCATED BEACHROCK MICROBIAL MATS1

Abstract: Photosynthetic electron transport of beachrock microbial mats growing in the intertidal zone of Heron Island (Great Barrier Reef, Australia) was investigated with a pulse amplitude modulation chl fluorometer providing four different excitation wavelengths for preferential excitation of the major algal groups (cyanobacteria, green algae, diatoms/dinoflagellates). A new type of fiberoptic emitter-detector unit (PHYTO-EDF) was used to measure chl fluorescence at the sample surface. Fluorescence signals mainly ori… Show more

“…Fluorescence signals from four wavelengths (665,645, 520 and 470 nm) were deconvoluted to the two algal groups by inserting the best fitting reference spectra from the test species, using the Phyto-PAM software Phyto-WIN v1.47. Fluorescence parameters used (Fo, F,) were as described by Schreiber et al (2002).…”

“…When light was applied during burial, both F and F m ' increased, suggesting a state transition from State 2 to State 1 (Schreiber et al 1995, Campbell et al 1998, Schreiber et al 2002. Once unburied, both fluorescence yields (F and F m ') of cyanobacteria decreased prior to a continued decrease in F ' and an increase in F m '.…”

“…Rubisco activase; Salvucci et al 1985, Salvucci & Ogren 1996, Jensen 2000, also contributing to induction of photochemistry. These above processes have been used to explain the reactivation of desiccated beach-rock microbial mats after rehydration (Schreiber et al 2002).…”

“…Variable chlorophyll fluorescence is a non-invasive tool that has been used previously to monitor PSII 95 activity in desiccated organisms (Huppertz et al 1990;Schreiber et al 2002). When a dark-adapted sample is 96 illuminated, the fluorescence yield shows a characteristic induction of fluorescence emission, known as the 97 "Kautsky" curve.…”
