Application of a radiometric ear assay for studies of adjuvant arthritis in rats

Abstract: A radioisotopic method, originally developed for measuring the cellular response in delayed hypersensitivity lesions in mice, has been evaluated in adjuvant arthritic rats. Focal accumulation of 5-iodo-2'-de~xyuridine-'~'I (""IUdR) at a site of antigen challenge (left pinna) was measured and expressed as increased radioactivity in the challenged (left) over the unchallenged (right) ear (L/R ear ratio). Immunologic specificity of the assay was established with anti-lymphocyte globulin (ALG)-treated arthritic ra… Show more

“…NS: not significant. adjuvant arthritis is not yet clearly under stood, the development of arthritis is con sidered to be a T-cell mediated delayed-type hypersensitivity reaction to mycobacterial fragments (20,21). Therefore, the data indicate that cholesterol feeding considerably affects the immune function.…”

“…The development of arthritis is thought to be a T-cell mediated delayed-type hypersensitivity reaction [27][28][29][30] (in this respect it is analogous to that thought to occur in rheumatoid arthritis i31[) resulting from (1) activation of a latent, as yet unidentified, arthritogenic virus or pleuropneumonia (Mycoplasma) organism, (2) formarion of an immunogenic component of host tissue (i.e. due to the effects of the adjuvant), or (3) combination of these two processes [32,33].…”

“…The present experiments also demon strated that adjuvant treatment in addition to cholesterol feeding is essential for causing cholesterol accumulation in the aorta. The development of adjuvant arthritis in the rat is thought to be a cell-mediated delayed type hypersensitivity reaction to mycobacterial fragments (24,25). Also, immunological injury to the vessel wall promotes athero sclerosis, as has been shown by the induction of serum sickness in the rat (3).…”
