Due to global ecological and economic challenges that have been correlated to the transition from fossil-based to renewable resources, fundamental studies are being performed worldwide to replace fossil fuel raw materials in plastic production. One aspect of current research is the development of lignin-derived polyols to substitute expensive fossil-based polyol components for polyurethane and polyester production. This article describes the synthesis of bioactive lignin-based polyurethane coatings using unmodified and demethylated Kraft lignins. Demethylation was performed to enhance the reaction selectivity toward polyurethane formation. The antimicrobial activity was tested according to a slightly modified standard test (JIS Z 2801(JIS Z :2010. Besides effects caused by the lignins themselves, triphenylmethane derivatives (brilliant green and crystal violet) were used as additional antimicrobial substances. Results showed increased antimicrobial capacity against Staphylococcus aureus. Furthermore, the coating color could be varied from dark brown to green and blue, respectively.Coatings 2019, 9, 494 2 of 16 and DNA in cells of mice against oxidation damage [19] have been tested. Gao [20] and Bshena [21] studied the antimicrobial activity of various textiles, using lignin incorporated into polyethylene films and applied in the finishing processes. For textiles, there are special requirements such as non-toxicity to the consumer, namely cytotoxicity, allergy or irritation and sensitization. In other recent studies, lignosulfonic acid is reported to exhibit broad-spectrum anti-HIV (human immunodeficiency virus) and anti-HSV (herpes simplex virus) properties [22,23]. Thus, Qiu investigated the anti-HIV-1 activity-potential of lignosulfonates as a microbicide to prevent HIV-1 sexual transmission [23]. Another recently reported study revealed that the antimicrobial capacity of lignin correlates with the phenolic components, specifically the side chain structure and the nature of further functional groups [24]. Typically, the presence of a double bond in α, β positions of the side chain and a methyl group in the γ position grants the phenolic fragments with the most potency against microorganisms. However, none of the hitherto published studies included the investigation of the antibacterial activity of lignin when included in polymeric matrices.Unmodified lignin is widely studied as a component for polymer production with a focus on phenol-formaldehyde resins and polyurethanes (PUs) [25], where lignin is used as polyol substitute due to the high amount of hydroxyl groups resulting in high crosslinking densities and variable mechanical properties [26][27][28]. Hybrid and antimicrobial nanoparticles (NPs) of poly (methyl methacrylate) (PMMA) in the presence of poly (diallyl dimethyl ammonium) chloride (PDDA) were previously obtained by emulsion polymerization in absence of surfactant with low conversion. In the presence of amphiphiles such as cetyl trimethyl ammonium bromide (CTAB), dioctadecyl dimethyl ammonium bromide (DODAB) or soybean lecithin, we found that conversion increased substantially. In this work, the effect of the amphiphiles on the NPs core-shell structure and on the antimicrobial activity of the NPs was evaluated. NPs dispersions casted on silicon wafers, glass coverslips or polystyrene substrates were also used to obtain antimicrobial coatings. Methods for characterizing the dispersions and coatings were based on scanning electron microscopy, dynamic light scattering, determination of thickness, rugosity, and wettability for the coatings and determination of colony-forming unities (log CFU/mL) of microbia after 1 h interaction with the coatings or dispersions. The amphiphiles used during PMMA/PDDA/amphiphile NPs synthesis reduced the thickness of the NPs PDDA shell surrounding each particle. The antimicrobial activity of the dispersions and coatings were due to PDDA—the amphiphiles were either washed out by dialysis or remained in the PMMA polymeric core of the NPs. The most active NPs and coatings were those of PMMA/PDDA/CTAB—the corresponding coatings showed the highest rugosity and total surface area to interact with the microbes. The dispersions and coatings obtained by casting of the NPs dispersions onto silicon wafers were hydrophilic and exhibited microbicidal activity against Escherichia coli, Staphylococcus aureus, and Candida albicans. In addition, a major effect of reduction in particle size revealed the suitability of nanometric and cationic NPs (sizes below 100 nm) represented by PMMA/PDDA/CTAB NPs to yield maximal microbicidal activity from films and dispersions against all microbia tested. The reduction of cell viability by coatings and dispersions amounted to 6–8 logs from [PDDA] ≥ minimal microbicidal concentration.

Biofilms located within the root canals of teeth are a unique and pressing concern in dentistry and in medical microbiology. These multispecies biofilms, which include fungi as well as bacteria, form in a protected site with low shear stress and low oxygen tension. Systemic antibiotics are of limited value because of the lack of blood flow of the site, and issues with innate and acquired resistance. Physical disruption using hand or rotary powered instruments does not reach all locations in the root canal system where biofilms are present. Alternative strategies including agitated irrigation fluids, continuous chelation, materials with highly alkaline pH, and antimicrobial nanoparticles are being explored to meet the challenge. Detection and quantification of biofilms using fluorescence-based optical methods could provide an indication of successful biofilm removal and an endpoint for physical and chemical treatments.
