Application of Doehlert Experimental Design for the Optimization of Medium Constituents for the Production of L-asparaginase from Palm Kernal Cake (Elaeis guineensis)

Abstract: Doehlert experimental design (DD) was applied for the optimization of medium constituents for L-asparaginase production by Yarrowia lipolytica NCIM 3472 in solid state fermentation (SSF) using palm kernel cake as the substrate. From the results of preliminary experimental runs, the three variables (glucose, moisture content, L-asparagine) have been identified as the potential variables for the production of L-asparaginase. Fifteen experimental runs designed by DD are carried out and the process response is mod… Show more

“…organic acids, enzymes (e.g. lipases and esterases [30], phosphatases [31], asparaginases [32], laccases [33], inulinase [34], mannosidase [35], single-cell proteins [36] and single-cell oils [37]. Furthermore, Y. lipolytica is oleaginous yeast species able to accumulate significant amounts of lipids in biomass and is considered to be an outstanding producer of lipids and has repeatedly been used as a model microorganism for fatty acid metabolism and lipid biosynthesis by ex novo route [38,39], which is characterized by incorporation of intermediates or final products of β-oxidation into triacylglycerol molecules accumulated in the lipid bodies of yeast cells [24].…”

“…Malaysia is emerging as one of the leading biofuel producers, with 91 palm oil plants approved and a handful now in operation [72]. The global production of Palm Kernel Cake (PKC), a by-product of oil extraction from the nut of the palm tree, Elaeis guineensis, is ever increasing due to the tremendous growth of the oil palm industry in many parts of Asia and Africa (PORLA, 2000) [73].…”

“…The Doehlerts experimental design is one of the response surface designs that is used to define optimum process parameters. Optimization employing the Doehlert experimental design is usually carried out in the following classical steps: experimentation, modeling, or determination of model coefficients, response prediction, and model validation (Karanam & Medicherla, 2010). The Doehlert's design has the ability to significantly reduce the number of runs required for the evaluation of the effects of experimental variables and their interactions, which as a consequence, reduces cost and time required for the process thereby rendering the whole process economically efficient (Ferreira et al, 2007).…”
