Application of metabolomics to prostate cancer

Abstract: The prostate has long been known to exhibit unique metabolite profiles. In the last decade, advances in nuclear magnetic resonance spectroscopy and mass spectrometry have been applied toward identifying metabolic alterations in prostate cancer that may provide clinically useful biomarkers. As with genomics and proteomics, advances in technology and bioinformatics have led to the application of metabolomic profiling to prostate cancer – the high throughput evaluation of a large complement of metabolites in the … Show more

“…Since the initial demonstration that sarcosine abundance is associated with prostate cancer aggressiveness, several investigators have worked to determine how best to apply metabolomics in cancer diagnosis and patient management, reviewed in Refs. [41,42].The present study examined over 500 prostate tissue samples from two independent cohorts of patients, using a state of the art metabolomics platform comprised of UHPLC-MS/MS and GC-MS [27,28]. This study identified biochemical correlates of prostate cancer aggressiveness using a combination of surgical and pathological findings, namely Gleason pattern, extracapsular extension, and spread to seminal vesicles and/or lymph nodes.…”

“…However, efforts at profiling the entire measured metabolome rather than focusing on individual metabolites have been increased 101 . Thus, a more complete depiction of the metabolic alterations among the development of cancer cells could be achieved 102,103,104,105 .…”

“…When nuclei in a magnetic field are exposed to a radiofrequency pulse their protons temporarily move to a higher energy state, and then release a characteristic radiowave when they return to their normal energy state [75]. The resulting NMR spectrum is a collection of characteristic peaks and intensities of each compound that allow its identification [76].…”
