Application of ultra performance liquid chromatography-photodiode detector-quadrupole/time of flight-mass spectrometry (UPLC-PDA-Q/TOF-MS) method for the characterization of phenolic compounds of Lepidium sativum L. sprouts

Abstract: Some of health-protecting phytochemicals can be found in the sprouts in a much higher concentration than in the developed plants. However, the polyphenol composition of Lepidium sativum L. sprouts is still unresolved, especially with regard to flavonoids and phenolic acids. To develop a rapid and reliable UPLC-PDA-Q/TOF-MS method for the identification and quantification of main flavonoids and phenolic acids from L. sativum sprouts, the samples of L. sativum sprouts were grown with and without the exposition t… Show more

“…The differential activities of flavonoids could depend on the solvent used for extraction and on the position of substituents in the flavonoid nucleus. These differences in substituents give rise to the major bioactive compounds such as quercetine, kampferol and their glycosides which are present in Lepidium sativum seeds [7,[15][16][17].…”

“…Sinapine was found to be the most dominant phenolic within the RSP extract at 10.91 ± 0.97%. Preliminary MS scans ( Figure S1) of the extracts had also shown the presence of other compounds which were putatively identified based on literature (Clauβ et al, 2011;Ferreres et al, 2009;Oszmiański, Kolniak-Ostek, & Wojdyło, 2013;Thiyam et al, 2009;Yang et al, 2015) as derivatives of sinapic acid or benzoylcholine, cyclic spermidine conjugate and feruloyl choline (5-8') guaiacyl (Table S2). Due to unavailability of authentic standards, we have not quantified these additional compounds.…”

“…The identification and quantification of the polyphenols was carried out with the use of an ACQUITY Ultra Performance LC system equipped with photodiode array detector with a binary solvent manager (Waters Corporation, Milford, MA, USA) series with a mass detector G2 Q/TOF micro mass spectrometer (Waters, Manchester, U.K.) equipped with an electrospray ionization (ESI) source operating in negative and positive modes [ 54 ]. Separations of individual polyphenols were carried out using a UPLC BEH C18 column (1.7 µm, 2.1 mm × 100 mm; Waters Corporation, Milford, MA, USA) at 30 °C.…”
