“…As a matter of fact there was always the danger of sensitising the patient towards the animal protein with all the unwanted consequences such as anaphylactic shock in all its forms. These effects were seen indeed in about five percent of people repeatedly undergoing this therapy (1,2). For those reasons an iatrogenically inactivated adrenal cortex was often preferred as presenting a less serious risk than that accompanying regular administration of non-human protein hormone.…”

“…On account of the above advantages corticotropin has been increasingly used in many disease states requiring long-term therapy with corticosteroids (9, 13,16). However, the repeated injections of corticotropin of animal origin, a straight chain of 39-amino acids, frequently results in antibody formation, and its wide clinical application has been limited on account of serious allergic reactions (3,4,10,14,17,20) and the occasional development of acquired resistance (19). The sensitization is probably related to contaminated proteins and/or to N-25-N-33 species specific amino acid sequence (6).…”

“…A compound identical with the first 24 amino acids of corticotropin bas been recently synthesized by Kappeler & Schwyzcr (12) possessing little immunologic although full steroidigenie activity (7, ir, 18). This synthetic polypeptide-j8^-^' corticotropin seems to be free of allergic reactions, even in patients allergic to ACTH of animal origin (3,5,14). The depot form of this tetracosapeptide (Depot-Synacthen*) is now being studied and in tbis paper tbe clinical efficacy in asthmatic patients of its intermittent long-term administration is described.…”
