Aptamer Directly Evolved from Live Cells Recognizes Membrane Bound Immunoglobin Heavy Mu Chain in Burkitt's Lymphoma Cells

Abstract: The identification of tumor related cell membrane protein targets is important in understanding tumor progression, the development of new diagnostic tools, and potentially for identifying new therapeutic targets. Here we present a novel strategy for identifying proteins that are altered in their expression levels in a diseased cell using cell specific aptamers. Using an intact viable B-cell Burkitt's lymphoma cell line (Ramos cells) as the target, we have selected aptamers that recognize cell membrane proteins… Show more

“…The proteins cross-linked to the biotinylated TAs were collected by binding them to streptavidin magnetic beads and the proteins were subsequently proteolyzed on the beads. The resulting peptides were analyzed by mass spectrometry (10)(11)(12), which identified annexin A2 as the likely binding partner for both Endo28 and Endo31 (Supplemental Tables 3-5).…”

“…These TAs (50 nM) were tested for their relative binding affinity for ECs on ovarian cancer tissue sections. Annexin A2 protein was identified as the target of these selected TAs by cross-linking and mass spectrometry (12).…”

“…Also, this method is further applied to cells to find cell-specific aptamers using the cell-SELEX method. Compared with other affinity tags, aptamers have many advantages, including small size, non-toxicity, relatively easy preparation and functionalization with no batch-to-batch variations and easy surface immobilization via their functional groups [51][52][53][54][55]. Thus, they can be easily functionalized simultaneously with (-SH) sulfhydryl groups and fluorescent dyes (fluorophores) to be tethered on the surface of AuNRs for both targeting and imaging purposes.…”

“…The identification of identity of aptamer target was achieved by chemically modifying the aptamers with a photoactive uracil derivative for covalent binding of aptamer with their targets. Once aptamers are bound to targets covalently, they are isolated through magnetic extraction and the target protein was determined through mass spectroscopy techniques (Mallikaratchy et al, 2007).…”
