Aptamer-Modified Erythrocyte Membrane-Coated pH-Sensitive Nanoparticles for c-Met-Targeted Therapy of Glioblastoma Multiforme

Abstract: Biomimetic drug delivery systems, especially red blood cell (RBC) membrane-based nanoparticle drug delivery systems (RNP), have been extensively utilized in tumor drug delivery because of their excellent biocompatibility and prolonged circulation. In this study, we developed an active targeting pH-sensitive RNP loaded with DOX by decorating an aptamer SL1 on RBC membranes (SL1-RNP-DOX) for c-Met-targeted therapy of glioblastoma multiforme (GBM). SL1 could specifically bind to c-Met, which is highly expressed i… Show more

“…These ligand‐receptor interactions lead to internalization of the aptamer‐containing molecule 115 . This phenomenon has been beautifully demonstrated by Liu and his team, who designed DOX‐loaded, pH‐sensitive PLGA NPs coated with aptamer SL1‐modified red blood cell membranes (RNPs) 116 . SL1 binds specifically to c‐Met, a protein found at high concentrations in GBM U87MG cells, thus enabling the accumulation of DOX in the tumor cells.…”

“…In this regard, Ben-Akiva et al synthesized PLGA NPs in anisotropic shapes and coated them in the Red blood cells (RBCs) membrane for more extended blood circulation ( Ben-Akiva et al, 2020 ). In GBM, CM-coated PLGA has been loaded with DOX or docetaxel (DTX) for enhanced chemotherapy in the GBM tumor site ( Chai et al, 2017 ; Zou et al, 2018 ; Liu et al, 2022b ).…”

“…In GBM, RBC is a popular choice for therapeutic agent encapsulation. For instance, RBC membrane-coated NPs have been designed for the delivery of therapeutic agents, including famous DOX, PLGA ( Chai et al, 2017 ; Ben-Akiva et al, 2020 ; Liu et al, 2022b ), gene therapy agents like DNA and RNA ( Liu et al, 2020a ; Han et al, 2021 ; Li et al, 2022a ), and agents like indocyanine green (ICG) for synergic GBM therapy ( Wang et al, 2020 ; Li et al, 2022a ). RBC membrane-coated NPs could also be used for toxin removal alongside cancer therapy.…”
