Aptasensors for the detection of infectious pathogens: design strategies and point-of-care testing

Abstract: The epidemic of infectious diseases caused by contagious pathogens is a life-threatening hazard to the entire human population worldwide. A timely and accurate diagnosis is the critical link in the fight against infectious diseases. Aptamer-based biosensors, the so-called aptasensors, employ nucleic acid aptamers as bio-receptors for the recognition of target pathogens of interest. This review focuses on the design strategies as well as state-of-the-art technologies of aptasensor-based diagnostics for infectio… Show more

“…MNPs with proper surface functionalization are especially suitable for the development of lab-in-chip biosensors for easy, reliable, and cost-effective POC diagnostic facilities. In addition, MNPs are widely used in the molecular diagnosis of human viruses [ 128 ] and other infectious pathogens [ 129 ]; MNP-based biosensors are also an extremely promising form in highly accurate POC SARS-CoV-2 detection [ 130 , 131 , 132 ]. In this review, we briefly highlight the following biological objects, which have been recently analyzed using MNP-based POC devices: SARS-CoV-2, Human chorionic gonadotropin (hCG), the typical oral pathogen of Streptococcus, foodborne bacteria in food and environmental samples ( Listeria monocytogenes , Campylobacter jejuni , and Staphylococcus aureus ), pathogenic bacteria ( Escherichia coli ( E. coli ), and Staphylococcus aureus ( S. aureus ), the food pathogen Vibrio parahaemolyticus ), the bacteria Acinetobacter baumannii , and influenza A H1N1 virus ( Table 1 ).…”

“…By using this method, specific interactions between the target bacterium and the recognition molecule on the QCM chip surface could be created for the quick determination of foodborne pathogens in food samples. Although many methods have been reported to detect pathogens in food samples, nonspecific interactions hinder real-life applications of sensor systems [ 19 , 25 – 27 ]. Therefore, the magnetic pre-enrichment technique has been used to prevent the interaction of impurities with the QCM system for the isolation of protein, cells, and fine chemicals from complex solutions without using expensive laboratory equipment.…”

“…From the point of view of medical diagnostics, assay accuracy as well as sensitivity and specificity (defined in biomedical terms) play a key role. The capability of the determination of trace concentrations of analytes, which enable the detection of various disease markers (e.g., the presence of viruses or bacteria) as well other abnormalities of the human body (usually manifested by the elevated levels of specific biomarkers), harmful chemical contamination in the environment, or poor-quality food, is also of great importance in various areas of life [ 2 , 3 , 4 , 5 , 6 ]. An important branch of modern analytical devices in this area is chemical sensors, which, still strive for the lack of sufficient sensitivity and selectivity in the case of the analysis of complex samples.…”
