Aqueous Blackcurrant Extract Improves Insulin Sensitivity and Secretion and Modulates the Gut Microbiome in Non-Obese Type 2 Diabetic Rats

Abstract: This study was undertaken to determine whether aqueous blackcurrant extracts (BC) improve glucose metabolism and gut microbiomes in non-obese type 2 diabetic animals fed a high-fat diet and to identify the mechanism involved. Partially pancreatectomized male Sprague–Dawley rats were provided a high-fat diet containing 0% (control), 0.2% (L-BC; low dosage), 0.6% (M-BC; medium dosage), and 1.8% (H-BC; high dosage) blackcurrant extracts; 0.2% metformin (positive-C); plus 1.8%, 1.6%, 1.2%, 0%, and 1.6% dextrin, sp… Show more

“…Thus, the animal model in the present study progressed from the acute stage of memory deficit to Alzheimer’s disease-like symptoms. Scopolamine is also reported to increase oxidative stress in the brain, and bioactive components to reduce oxidative stress, such as flavonoids (apigenin and silymarin), prevent memory impairment by scopolamine [ 22 , 23 ]. Their intake reduces reactive oxygen species and increases glutathione (GSH), similar to donepezil.…”

“…The metabolic functions of the gut microbiota were predicted from the fasta files and count tables of fecal bacteria using PICRUSt2 [ 22 ]. The metabolic functions were based on the Kyoto Encyclopedia of Genes and Genomes (KEGG) Orthologues (KO) mapped using KEGG mapper ( , accessed on 11 November 2021) [ 41 ].…”

“…β-glucan is one of the prebiotics that decreases serum glucose concentration with increasing propionate and butyrate-producing bacteria in the gut [ 41 ]. Anthocyanins like Aronia and black currant also promote glucose metabolism by increasing the relative abundance of Bacteroides , Prevotella , and Akkermansia to increase butyrate production [ 13 , 42 , 43 ]. Therefore, AGM intake might promote gut microbiota composition contributing to improving glucose metabolism.…”
