“…A GST pull‐down assay was conducted as described in [29]. Briefly, crude extract of GST‐AtTrx m2 fusion protein was bound to Glutathione Sepharose 4 Fast Flow (GE Healthcare) according to the manufacturer's instructions.…”

“…To identify AGB1-interacting proteins, we used AGB1 as a bait protein for a yeast two-hybrid screen of a normalized Arabidopsis cDNA library (for details, see Methods). The screening identified two potential AGB1 interactors: NPH3, a regulator of hypocotyl phototropism (Kansup et al, 2014), and BBX21 (GenBank: AAO42831; AT1G75540), a BBX-containing transcription factor. Because BBX21 is known to play a vital role in hypocotyl elongation (Datta et al, 2007;Xu et al, 2016), we selected BBX21 for further analysis.…”

“…Hypocotyl development in Arabidopsis thaliana is a delicate developmental process regulated through the complex interplay between light and endogenous hormonal cues (Neff et al, 2006;Alabadí and Blá zquez, 2009;Lau and Deng, 2010;Boron and Vissenberg, 2014). Two mutants in AGB1 (agb1-1 and agb1-2) have a short hypocotyl phenotype and display decreased hypocotyl phototropism, relative to wild-type (Ullah et al, 2003;Chen et al, 2004;Kansup et al, 2014). In contrast, gpa1-4 (GPA1) has a slightly short hypocotyl phenotype, and gcr1-2 (GCR1, G-Protein-Coupled Receptor 1) hypocotyls develop normally compared with wild-type (Chen et al, 2004).…”

“…To date, Y2/3H screening has been the primary method used to identify AGB1 interactors 125 (Mudgil et al, 2009;Kobayashi et al, 2012;Tsugama et al, 2012;Kansup et al, 2013;126 Tsugama et al, 2013;Kansup et al, 2014;Xu et al, 2015). In order to identify AGB1-associated 127 proteins in planta, we performed co-IP from transgenic Arabidopsis expressing 35S driven 128 FLAG-tagged AGB1 in the agb1-2 background, or from wild-type plants as a negative control.…”
