Arabidopsis Phyllotaxis Is Controlled by the Methyl-Esterification Status of Cell-Wall Pectins

Abstract: Plant organs are produced from meristems in a characteristic pattern. This pattern, referred to as phyllotaxis, is thought to be generated by local gradients of an information molecule, auxin. Some studies propose a key role for the mechanical properties of the cell walls in the control of organ outgrowth. A major cell-wall component is the linear alpha-1-4-linked D-GalAp pectic polysaccharide homogalacturonan (HG), which plays a key role in cell-to-cell cohesion. HG is deposited in the cell wall in a highly (… Show more

“…The regulation of the DM of HG has been implicated in many aspects of plant development (Wolf et al, 2009a), including cell adhesion (Tieman and Handa, 1994;Wen et al, 1999;Mollet et al, 2000;Bouton et al, 2002;Mouille et al, 2007;Durand et al, 2009), adventitious rooting (Guénin et al, 2011), primordia emergence at the shoot apical meristem (Peaucelle et al, 2008(Peaucelle et al, , 2011, fruit ripening (Brummell and Harpster, 2001;Phan et al, 2007), and plant defense (Bethke et al, 2014). Although data have shown an important role for Figure 5.…”

“…Because the degree of HG methylation affects the stiffness of the cell wall (Peaucelle et al, 2008), the LM20 and JIM7 antibodies were used to look at the highly methylesterified HG distribution ( Figure 5; Supplemental Figure 8). An increase in LM20 labeling in AM was observed in the uuat1-2 mutant when compared with wild-type Col-0 (Figures 5A and 5B), suggesting the (A) Immunoblot of the yeast microsomal fractions used to make the proteoliposomes.…”

“…Four PMEIs have been functionally characterized in Arabidopsis to date. PMEI1 and PMEI2 (Wolf et al, 2003) are highly expressed in flowers; PMEI1 expression is specific to pollen, while the expression of PMEI3 is particularly high in the apical meristem (Peaucelle et al, 2008). PMEI4 is up-regulated during growth acceleration in skotomorphic hypocotyls (Pelletier et al, 2010).…”
