Architecture of African swine fever virus and implications for viral assembly

Abstract: African swine fever virus (ASFV) is a giant and complex DNA virus that causes a highly contagious and often lethal swine disease for which no vaccine is available. Using an optimized image reconstruction strategy, we solved the ASFV capsid structure up to 4.1 angstroms, which is built from 17,280 proteins, including one major (p72) and four minor (M1249L, p17, p49, and H240R) capsid proteins organized into pentasymmetrons and trisymmetrons. The atomic structure of the p72 protein informs putative conformationa… Show more

“…The recent 'resolution revolution' in cryo-EM has yielded high-resolution structures of a wide array of previously intractable biological systems (Yuan et al, 2018;Zhu et al, 2018;Wang et al, 2019). In the current paper, the high resolution cryo-EM structures succinctly revealed six copies of the complex.…”

“…Due to the lack of vaccine and effective treatments against ASFV, disease control mainly relies on culling pigs 1,4 . More than 30 million domestic pigs were culled in the past 2 years, and the number continues to increase 6 . Thus, rapid diagnosis of the ASFV-affected animal is crucial to prevent its broad expansion, and to reduce economic losses 7,8 .…”

“…ASFV is the only member of the Asfarviridae family. Prior studies showed that the ASFV virion has a complex structure with multiple membrane and protein layers [5,6]. The major capsid protein p72, one of the key protective antigens, form homo-trimers and assemble the outmost icosahedral protein shell [6,7].…”

“…Prior studies showed that the ASFV virion has a complex structure with multiple membrane and protein layers [5,6]. The major capsid protein p72, one of the key protective antigens, form homo-trimers and assemble the outmost icosahedral protein shell [6,7]. The structure of p72 trimer was determined by Liu, Q., et al [7], their result also revealed that p72 molecules can be assembled to form a thermostable trimer when B602L was co-expressed with p72; but the structure of B602L was remain unknown.…”
