Are Ionic Liquids Better Extracting Agents Than Toxic Volatile Organic Solvents? A Combination of Ionic Liquids, Microwave and LC/MS/MS, Applied to the Lichen Stereocaulon glareosum

Abstract: We report a green strategy for the extraction of lichen substances from Stereocaulon glareosum. This sustainable alternative does not use volatile toxic organic solvents, but it is assisted by microwave and is checked by UHPLC/ESI/MS/MS. Ionic liquids may provide a better alternative in the extraction of natural products from lichens.

“…22 Moreover, the interest of these cations to extract various lichen metabolites was recently confirmed. 23 We have also reported an optimisation study 27 using six imidazolium and pyridinium-based ILs for MAE from Pseudevernia furfuracea. Microwave irradiation is generally more efficient to extract the targeted lichen metabolites (Figure 1) than conventional heating.…”

“…22 An additional study was recently performed on Stereaucolon glareosum confirming the ability of imidazolium cations to interact with lichen metabolites. 23 In this work, we explore various parameters influencing the IL-MAE of four metabolites from Pseudevernia furfuracea var. furfuracea (Figure 1) including the method used to prepare the samples and the structure of ILs.…”

“…For example, for the extraction of non-polar components, such as lipids, polyketides and terpenoids, one may use water-immiscible solvents such as ethyl acetate or dichloromethane. Other water-immiscible solvents such as ethers, chloroform and carbon tetrachloride are less common due to their toxicity and negative environmental effects [134,179]. Additionally, pH is crucial for the extraction, due to ionizable moieties in the chemical structure of the metabolites being extracted into the organic phase to a much higher degree in their neutral form than in a charged state.…”

“…Other chromatographic techniques use two consecutive separation columns (2-dimensional liquid chromatography) or columns of different sorbent chemistry, such as ionic exchangers (see Table 1) [14,113,129,134,143]. The most common ionization techniques used in combination with 2D liquid chromatography are the ambient pressure ionization techniques, such as electrospray ionization (ESI) [134,135,137,138,[140][141][142][143][144][145]150] and atmospheric pressure chemical ionization (APCI) [139,146]. These are all soft ionization techniques (i.e., they produce limited chemical fragmentation).…”

“…New alternative solvents have gained increasing attention in research due to the negative impact of traditional chemicals on people and the environment. Therefore, they are employed as eco-friendly extractants and sorbents to replace the old ones (Sepulveda et al, 2020). Mainly, ionic liquids (ILs) and deep eutectic solvents (DESs) contribute greatly to separation science besides their use as an extraction medium (Marták and Schlosser, 2019); they can also assist adsorbents to improve separation performance by the disaggregation of nano-materials into smaller bundles (Zaib et al, 2020).…”
