Asp f6, an Aspergillus allergen specifically recognized by IgE from patients with allergic bronchopulmonary aspergillosis, is differentially expressed during germination

Abstract: Aspergillus fumigatus is able to colonize its environment by the formation of hyphae. Hyphae are found in the lung of ABPA patients, but not in patients suffering from atopic asthma. Our finding that Asp f6 is specifically expressed in hyphae might explain why an IgE response to Asp f6 is specific for ABPA patients.

“…7A, B, and D). To determine whether the amorphous material at the hyphal tips is released cytoplasm, we stained ⌬afmnt1 samples using the monoclonal antibody B8-C4, which specifically recognizes an epitope in the conserved domain of the acid ribosomal proteins P0, P1, and P2 (29). Immunofluorescence analysis revealed a strong labeling of the amorphous material at the hyphal tips of the ⌬afmnt1 mutant (Fig.…”

“…Yet, it is quite surprising that reliable methods for the extraction of proteins from filamentous fungi did not emerge until the early 2000s. Since then, we, and others, have reported essentially similar strategies for mycelial lysis involving vigorous physical disruption (bead-beating [19], grinding in liquid N 2 [7,20], sonication or the use of French press technology [21]). Despite the availability of efficient approaches to cell wall disruption, it is only recently that a strategy for the identification of intracellular proteins from A. fumigatus has been forthcoming [22].…”

“…30 The r Asp f6 antigen has been identified as a protein specific to the hyphae of Aspergillus, and these hyphae are only present in ABPA. 31 Therefore, identification of IgE specific to the r Asp f4 and r Asp f6 antigens can also be used as a marker specific to ABPA. 30 In our sample, and r Asp f6, and an association with total serum IgE over 1,000 UI/ml allowed ABPA to be confirmed with high specificity (100%) and sensitivity (64%).…”
