Aspartic protease inhibitor enhances resistance to potato virus Y and A in transgenic potato plants

Abstract: Background Viruses are the major threat to commercial potato (Solanum tuberosum) production worldwide. Because viral genomes only encode a small number of proteins, all stages of viral infection rely on interactions between viral proteins and host factors. Previously, we presented a list of the most important candidate genes involved in potato plants’ defense response to viruses that are significantly activated in resistant cultivars. Isolated from this list, Aspartic Protease Inhibitor 5 (API5… Show more

“…The authors constructed a vector with the potato StAPI35 gene under the control of the cauliflower mosaic virus (CaMV) 5S promoter. Transgenic plants expressing StAPI5 demonstrated significant viral resistance in comparison to the control, indicating that the StAPI5 gene is imperative in enhancing plant resistance to viral stress ( Osmani et al, 2022 ). It is possible that the downregulation of certain plant defense genes during the BP may contribute to PVY infestation.…”

“…For this reason, several protease inhibitor drugs, including azithromycin, were tested in the last coronavirus pandemic, demonstrating a clear example of their increasing use in clinical medicine as antiviral agents [ 38 ]. Expression of the rice cysteine protease inhibitor gene oryzacystatin I in transgenic tobacco showed enhanced resistance to two potyviruses, tobacco etch virus and potato virus Y, which use cysteine proteases for protein processing [ 8 ]. Specific protease inhibitors are also expressed naturally in response to viral infections.…”
