Assay of Phospholipase A Activity

Abstract: Phospholipases of the A type constitute a large family of esterases that catalyze the hydrolysis of the fatty acid ester bonds in phospholipids and thus generate lysophospholipids and fatty acids. Both products or their metabolites are important signal molecules in the cellular adaptation to stress, developmental processes and several diseases in plants and animals. The assay of PLA activity has been much promoted by the availability of phospholipid substrates with fluorophores at one or two fatty acids. The d… Show more

“…Lysophospholipids and their metabolites are signaling molecules that are involved in cellular adaptation to stress, developmental processes, and various diseases in both plants and animals [ 93 ]. Current evidence suggests that PLA represents a crucial target involved in the modulation of intracellular molecular pathways, leading to the generation of lipid mediators, which control and regulate cell proliferation, survival, differentiation, motility, and tissue vascularization [ 94 ]. Therefore, the deregulation of PLA is involved in the pathogenesis of several diseases and cancer development and progression.…”

“…After adding a protein with phospholipase activity, the lipids will be cleaved, so one of the two acyl chains is cleaved off. This will create a distance between the two fluorescent molecules, and they will start to emit a detectable signal (Figure 12c) (52,53).…”

“…The RNAi vector DNA was fixed at gold particles and used for the biolistic bombardment of E. californica wild-type cells by the Bio-Rad PDS-1000/He Particle Delivery System, following a recently published protocol (Heinze and Roos, 2013). The stably transformed cell lines were grown over at least three 9-d passages on selection medium containing 100 mg mL 21 paromomycin and later maintained by growth cycles in medium with 50 mg mL 21 paromomycin.…”
