“…A low concentration is associated with high viscosity and high light-scattering signals. The weight-averaged molecular weight of HA ranged from 1 to 3 9 10 6 Da (but more generally approximately 1.5 9 10 6 Da), in agreement with previous results obtained with a similar technique [1,40]. This range of molecular weight values is lower than published results but similar to those obtained using other techniques [9,14,32].…”

“…Our assay gives a direct molecular weight determination by light scattering and determination of concentration by differential refractometry. A similar technique showed good agreement between SEC estimation after incubation with pronase to hydrolyze proteins [40]; direct determination by SEC seems to be a useful technique to determine molecular weight and concentration of HA in SF. The elution of HA in SEC is perfectly dissociated from that of the protein pool.…”

“…Identification was completed using a ultra-violet detector (UV) permitting us to conclude that proteins are (in most cases) completely separated from linear HA (Figure 2). These results confirm the data from literature obtained with only one different column (Superose 6 from Pharmacia [29] or TSK 6000PW from Toyo Soda [28,30]). …”

“…To determine the composition of SF, a relatively new technique was proposed [28]: steric exclusion chromatography (SEC) which separates the components following their hydrodynamic volume. Different conditions are indicated in the literature: direct injection after dilution [26,29,30] or dilution and incubation with pronase [26,31] before the SEC experiment, using a light scattering detector or calibration with standards. The influence of such protease pre-treatment before SEC analysis will be also examined in this paper.…”
